三药组合抑制由α-溶血素导致的巨噬细胞炎性因子激活

摘要 为了考察一种新型三药组合(氯唑西林、硫利达嗪和四环素)对金葡菌α-溶血素分泌的影响,以及探讨该三药组合对由金葡菌α-溶血素引起巨噬细胞炎症发生的抑制作用。试验采用抗菌药物敏感实验、三维棋盘法、溶血实验、Western blot实验方法。结果表明:三药组合显示出协同杀菌作用;三药组合能够显著协同抑制α-溶血素的分泌;三药组合可显著抑制由金葡菌α-溶血素引起巨噬细胞raw264.7相关炎症通路MAPKs、NF-κB、NLRP3蛋白的表达。说明该新型三药组合通过抑制金葡菌α-溶血素的分泌,进而阻断金葡菌α-溶血素引起的巨噬细胞炎性因子的激活。 To investigate the effect of a new three-drug combination(cloxacillin,thioridazine and tetracycline)on the secretion ofα-hemolysin from Staphylococcus aureus,and to explore the combination of the three drugs on the Staphylococcus aureusα-hemolysin inhibition of phagocyte inflammation.Antibacterial drug sensitivity test,three-dimensional chessboard method,hemolysis experiment,and Western blot experimental method were used.The results showed that the combination of three drugs showed synergistic bactericidal effects;the combination of three drugs could significantly inhibit the secretion ofα-hemolysin.The combination of three drugs could significantly inhibit the expression of MAPKs,NF-κB and NLRP3 proteins in macrophages raw264.7-related inflammatory pathway induced by Staphylococcus aureusα-hemolysin.This novel combination of three drugs inhibits the secretion of macrophage inflammatory factors caused by Staphylococcus aureusα-hemolysin by inhibiting the secretion of aureusα-hemolysin.
