漆酶催化偶氮染料酸性橙脱色的研究 被引量:3

摘要 利用米曲霉菌漆酶对偶氮染料酸性橙进行脱色实验,考察反应时间、pH、加酶量、温度、染料质量浓度等因素对脱色率的影响。结果表明,漆酶催化酸性橙的适宜条件为:反应时间为50 min、pH=6、加酶量为2 mL、反应温度为50℃、染料质量浓度为0.1 g/L,最适条件下酸性橙的脱色率达88.32%。同时研究了金属离子、酶抑制剂对酸性橙脱色的影响,结果发现十二烷基硫酸钠(SDS)显著抑制漆酶的催化活性,Fe^2+对漆酶脱色有完全抑制作用;Mg^2+对脱色有一定的促进作用。 Degradation of acid orange,which is a typical azo dye,is investigated by using laccase extracted from aspergillus oryzae.Influences of reaction time,p H value,adding amount of laccase,temperature,dye concentration on decolorization rate are explored.Besides,the decolorization effects of EDTA,SDS mediator and metal ions are studied.The suitable decolorization conditions for acid orange are found as follows:the reaction time is 50 minutes,pH=6,the enzyme dosage is 2 ml,the reaction temperature is at 50℃,and the initial dye concentration is 0.1 g·L^-1.Decoloration rate of acid orange is 88.32%under the most suitable conditions.The effects of metal ions and enzyme inhibitors on the decolorization of acid orange are also studied.It is found SDS can inhibit the catalytic activity of laccase significantly,Fe^2+has a whole inhibition to laccase,and Mg^2+has certain promoting effect on decolorization.
