四种去内毒素试剂盒提取质粒中内毒素水平的比较

A comparison of endotoxin levels in plasmids extracted by four kinds of endotoxin removal kits

目的比较四种去内毒素质粒提取试剂盒所提质粒中内毒素的水平。方法本实验选用了四种商业化去内毒素试剂盒进行质粒提取,然后用鲎试剂凝胶法和动态浊度法对所提质粒DNA进行内毒素检测。结果四种试剂盒所提取的质粒得率、A260/A280、A260/A230均符合要求,内毒素水平均小于0.1EU/μg。其中,C试剂盒所提取的质粒中内毒素含量最低。结论四种去内毒素质粒提取试剂盒在质粒得率、内毒素含量、价格方面有一定的差异。其中C试剂盒所提质粒得的率最高,内毒素含量最低,但价格也最贵;B试剂盒性价比最高,使用时应根据实际情况综合考虑。

Objective To compare endotoxin levels in plasmids extracted by four kinds of endotoxin-free plasmid extraction kit.Methods Four commercial endotoxin removal kits were selected for plasmid extraction in this experiment,and then Limulus reagent gel method and dynamic turbidity method were used for endotoxin detection of the plasmid DNA.Results The plasmid yield,A260/A280 and A260/A230 of the four kits all met the requirements,and the endotoxin level in each of the four cases was less than 0.1EU/μg.Among them,the plasmid extracted by kit C had the least endotoxin content.Conclusion There are some differences in plasmid yield,endotoxin content and price among the four kinds of endotoxin-free plasmid extraction kits.Among them,the plasmid extracted by kit C has the biggest yield,the least endotoxin content,but the highest price.Kit B has the highest cost performance and should be given comprehensive consideration according to the actual situation in its use.
