中药调控HIF-1α抑制肝癌的研究进展

Research progress on the inhibition of liver cancer of traditional Chinese medicine through the regulation of HIF-1α

缺氧是许多实体瘤的常见特征,可引起HIF-1α(缺氧诱导因子-1α)过度表达。HIF-1α是缺氧应答中关键的转录因子,其累积将导致肿瘤产生一系列恶性生物学行为。研究表明,HIF-1α在肝癌中高度表达,通过激活下游基因,使肝癌细胞异常增殖、凋亡耐受、肿瘤血管生成、能量代谢异常、侵袭和转移,与肝癌的发生发展有着密切联系。许多中药及其有效成分已被证实具有抗肝癌作用。因而本文总结了以HIF-1α为靶点的中药单体、中药提取物及中药复方对肝癌的影响,阐述了其主要作用机制,以期为进一步研究作为参考。

Hypoxia is the characteristic of many solid tumor,which can lead to the over expression of HIF-1α(hypoxiainducible factor-1α ).As the core factor involved in hypoxia regulation,HIF-1α will lead to a series of malignant biological behaviors.Studies have shown that HIF-1α is highly expressed in liver cancer.HIF-1α can induce abnormal proliferation,apoptosis tolerance,tumor angiogenesis,abnormal energy metabolism,invasion and metastasis of liver cells by inducing downstream gene expression,which has a close relationship with the occurrence and development of liver cancer.Various traditional Chinese medicine and their active ingredients have been confirmed to have anti-liver cancer effect.Therefore,the paper summarizes the effects of monomers,extracts and compounds prescription of traditional Chinese medicine on liver cancer,and explores the main mechanism in order to provide basis for further study.
