动物源性空肠弯曲菌的临床检测与特性分析 被引量:2

Clinical detection and characterization of Campylobacter jejuni isolates from animals

摘要 为做好畜禽疾病防控及动物性食品安全评价,根据空肠弯曲菌的16S rDNA基因和马尿酸酶基因(hipO)分别设计2对特异性引物,建立双重PCR方法用于空肠弯曲菌的检测。通过对参考菌株的测定,确定了其特异性高、灵敏度强,最小检出的DNA浓度达1 pg·μL-1。在采集的540份临床健康羔羊肛拭子样品和92份患病畜禽肠道样品中,共检出10份样品为空肠弯曲菌阳性,其中羊、鸡、鹅的阳性检测率分别为0.74%、10.5%和12.5%。通过细菌分离纯化,获得了10株空肠弯曲菌。体外培养试验发现羊源分离株生长速度低于鸡源和鹅源分离株。药敏试验表明所有分离株对磺胺类药物完全耐药;对头孢菌素类、四环素类、喹诺酮类药物的耐药性较高,且所有菌株均为多重耐药菌株。 To prevent the animal epidemic disease and evaluate the safety of meat food from animal sources,a duplex PCR method was performed by using two sets primers derived from the nucleotide sequence of 16S rDNA and the hippuricase gene(hipO)of Campylobacter jejuni.The high specificity and sensitivity of the PCR method were evaluated by using different bacteria,and the data showed the least concentration of DNA templates was 1 pg per microliter.Among the 540 samples from clinical healthy lambs and 92 samples from diseased sheep and poultry,10 samples were positive for C.jejuni.Thereinto,the samples from sheep were 0.74%positive,while the samples from chickens and geese were 10.5%and 12.5%respectively.Ten C.jejuni isolates were purified from the positive samples.The data of bacteria culture in vitro showed that C.jejuni isolated from sheep could grow faster than the isolates from chickens and geese.Drug susceptibility test indicated that all the 10 C.jejuni isolates were completely resistant to sulfonamides and highly resistant to Cephalosporins,Tetracyclines and Quinolones.Further more,all of bacteria isolates displayed multiple drug-resistant properties.
