孕康口服液对氢化可的松联合米非司酮致肾虚流产模型小鼠的影响 被引量:11

Effect of Yunkang Oral Liquid on Kidney-deficiency Abortion Model Miceinduced by Hydrocortisone Combined with Mifepristone

摘要 目的研究孕康口服液对肾虚流产模型小鼠的安胎作用,并观察其对性激素水平及Th1/Th2细胞因子的影响。方法将妊娠小鼠随机分为正常对照组、模型对照组、黄体酮组(39 mL·kg^-1)及孕康口服液低、中、高3个剂量组(9、18、36 mL·kg^-1)。采用灌胃给予氢化可的松(25 mg·kg^-1)与米非司酮(4 mg·kg^-1)复制肾虚流产小鼠模型,并同时灌胃给予孕康口服液连续9 d。给药期间测定小鼠背温及自主活动;取血测定血清中β-绒毛膜促性腺激素(β-HCG)、孕酮(P)、肿瘤坏死因子(TNF-α)、白细胞介素10(IL^-10)水平,并计算Th1/Th2细胞因子(TNF-α/IL^-10)比值;处死小鼠后,计算总胎数、死胎数与胚胎丢失率;取肾脏、卵巢,计算脏器指数,并进行肾脏和子宫组织形态学观察。结果与模型对照组相比,孕康口服液能升高模型小鼠背温(P<0.05,P<0.01)和自主活动(P<0.05),降低胚胎丢失率(P<0.01),升高卵巢指数(P<0.05,P<0.01),降低肾脏指数(P<0.05),升高血清P、β-HCG和IL^-10水平(P<0.05,P<0.01),降低血清TNF-α水平(P<0.05,P<0.01)及TNF-α/IL^-10比值(P<0.05,P<0.01),改善肾脏和子宫内膜病变情况。结论孕康口服液对氢化可的松联合米非司酮制备的肾虚流产模型小鼠具有明显的安胎作用,其作用机制可能与调节Th1/Th2细胞因子平衡及性激素水平有关。 Objective To study the anti-miscarriage effect of Yunkang oral liquid on the abortion of mice with kidney-deficiency and observe its effects on hormone levels and Th1/Th2 cytokines.Methods Pregnant mice were randomly divided into normal group,model group,progesterone(39 mL·kg^-1)group and Yunkang oral liquid low,medium,and high groups(9,18,36 mL·kg^-1).Preparation of mice abortion model with kidney-deficiency was carried out by intragastric administration of hydrocortisone(25 mg·kg^-1)combined with mifepristone(4 mg·kg^-1).Yunkang oral liquid was given at the same time for 9 consecutive days.The back temperature and spontaneous activity of mice during administration were measured;serum levels ofβ-HCG,P,TNF-α,IL^-10 were determined and the ratio of Th1/Th2 cytokines(TNF-α/IL^-10)was calculated.After the mice were sacrificed,the total number of fetuses,the number of stillbirths and the rate of embryo loss were recorded and calculated.The kidneys and ovaries were taken and the organ indexes were calculated.Morphological observation of kidneys and uterus were also performed.Results Compared with the model group,Yunkang oral liquid can increase the back temperature(P<0.05,P<0.01)and autonomous activity(P<0.05)of model mice;reduce abortion rate(P<0.01)and raise ovarian index(P<0.05,P<0.01);lower kidney index(P<0.05),increase serum levels of P,β-HCG and IL^-10(P<0.05,P<0.01);reduce serum TNF-αlevels(P<0.05,P<0.01)and TNF-α/IL^-10 ratio(P<0.05,P<0.01),improve kidney and endometrial lesions.Conclusion Yunkang oral liquid has obvious anti-miscarriage effect on kidney-deficiency abortion model mice induced by hydrocortisone combined with mifepristone,its mechanism may be related to the regulation of Th1/Th2 cytokine balance and sex hormone levels.
