从心论治方对ApoE^-/-小鼠血脂和肠道菌群的影响 被引量:6

摘要 目的探讨从心论治方对APOE基因敲除(ApoE^-/-)小鼠血脂和肠道菌群的影响。方法将36只雄性ApoE^-/-小鼠随机分为6组:模型组、对照组、辛伐他汀组,从心论治方高、中、低组。高脂饲料喂养12周制备高脂血症小鼠模型,进行药物处理;检测小鼠血脂水平;油红染色观察肝脏组织学变化;16s rDNA高通量测序检测小鼠肠道菌群组成和结构。结果模型组小鼠血脂水平明显升高,肝脏出现明显脂肪变性,肠道菌群结构紊乱。给药组小鼠总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)水平均降低(P<0.01,P<0.05),高密度脂蛋白胆固醇(HDL-C)水平升高(P<0.05),肝脏脂肪变性情况缓解。从心论治方组小鼠肠道菌群中厚壁菌门与拟杆菌门的比例降低,菌群多样性增加,肠道菌群失调得到改善。结论从心论治方能调节高脂喂养ApoE^-/-小鼠血脂水平,改善肝脏脂肪变性情况,改变肠道菌群组成结构。 Objective To investigate the effects of Congxinlunzhifang(CXLZF)on blood lipid and intestinal microbes in ApoE^-/-mice.Methods Thirty-six male ApoE^-/-mice were randomly divided into six groups:model group,control group,simvastatin group and three different doses of CXLZF groups.Mice were fed with high-fat diet for 12 weeks to prepare the hyperlipidemia models,and drug treatment was performed,blood lipid levels were measured;liver histological changes were observed by oil red staining;mice intestinal flora was detected by 16 s rDNA highthroughput sequencing.Results In the model group,the blood lipid levels of the mice were significantly increased,the livers of the mice showed obvious steatosis and the intestinal flora were disordered.Liver steatosis was relieved in the mice of the drug-administered groups;total cholesterol(TC),triglyceride(TG),low-density lipoprotein cholesterol(LDL-C)levels decreased(P<0.01,P<0.05),and high-density lipoprotein cholesterol(HDL-C)levels increased(P<0.05).In the CXLZF groups,the proportions of the Firmicutes and the Bacteroides in the intestinal flora of the mice decreased,the bacteria diversities increased and the intestinal flora imbalance were improved.Conclusion CXLZF can regulate blood lipid level of high-fat-fed ApoE^-/-mice,improve the liver steatosis and change the composition of intestinal flora.
