摘要
目的:观察对比剂肾病(contrast-induced nephropathy,CIN)大鼠真核翻译起始因子2α(eukaryotic initiation factors,eIF2α)的表达情况,探讨羟苯磺酸钙在对比剂肾病大鼠中的作用。方法:将42只大鼠随机分为3组:对照组(A组),模型组(B组),羟苯磺酸钙组(C组)。建立模型后24 h、48 h分批处死大鼠,观察各组大鼠肌酐(Scr)、尿素氮(BUN)、Cys C水平及肾脏病理改变,了解各组大鼠肾脏eIF2α的表达情况。结果:A组Scr、BUN、Cys C及eIF2α表达最低,且肾脏组织无明显病理变化。B组大鼠Scr、BUN、Cys C、eIF2α表达明显升高,差异有统计学意义(P<0.05),肾脏细胞凋亡显著。C组Scr、BUN、Cys C、eIF2α表达较B组明显降低(P<0.05),肾脏细胞凋亡明显减轻,但仍重于A组。结论:大鼠CIN的发生发展可能与内质网应激(endoplasmic reticulum stress,ERS)有关,羟苯磺酸钙在改善微循环的同时,可减轻内质网应激产生的不利影响。
Objective:To observe the expression of Eukaryotic translation initiation factors 2α(eIF2α)in contrast-induced nephropathy(CIN)rats and the intervention effects of calcium dobesilate for it.Methods:42 Wistar rats were randomly divided into 3 groups:Control group,CIN group and calcium dobesilate treated group.At different time points of 24 h,48 h after injection of contrast media,the rats were sacrified for tissue and blood collection.Blood urea nitrogen(BUN)serum creatinine(Scr)and Cys C were detected and pathological changes of kidney were evaluated,as well as the expression of renal eIF2α.Results:The expression of Scr,BUN,Cys C and eIF2αwas lowest in group A,and there were no obvious pathological changes in the kidney.Moreover,the expressions of Scr,BUN,Cys C and eIF2αin rats in group B were significantly increased,with statistically significant differences(P<0.05).The expressions of Scr,BUN,Cys C,eIF2αin group C were significantly reduced compared with group B(P<0.05),renal cell apoptosis was significantly reduced,but still more serious than group A.Conclusion:The occurrence and development of CIN in rats may be related to Endoplasmic Reticulum stress.Calcium dobesilate can play a protective role in reducing Endoplasmic Reticulum stress in rats with contrast nephropathy while improving microcirculation.
