响应面法优化半枝莲总黄酮微波提取工艺及其体外活性研究 被引量:8

Optimization of Microwave Extraction Technology of Total Flavonoids by Response Surface Methodology and in Vitro Activity Evaluation from Scutellaria Barbata

摘要 目的优化半枝莲总黄酮微波提取工艺,探讨其抗氧化活性及抗乙酰胆碱酯酶活性。方法以半枝莲为原料,在单因素试验基础上,采用响应面法考察提取次数、乙醇体积分数、料液比、微波提取时间、微波提取功率5个因素对总黄酮提取工艺的影响。同时利用二苯代苦味酰基(DPPH)自由基法、2,2-连氨-(3-乙基苯并噻唑啉-6-磺酸)二氨盐(ABTS)自由基法测定其自由基清除活性,采用改良的Ellman法评价抗乙酰胆碱酶活性。结果微波提取半枝莲总黄酮的条件为乙醇体积分数80%,微波提取功率525 W,料液比1∶24(mg/m L),微波提取时间13 min,实际得率为10.89%;半枝莲总黄酮提取物有良好的抗氧化活性及一定的抗乙酰胆碱酯酶活性。结论该方法稳定可行,可用于分析微波提取半枝莲总黄酮得率。 Objective To optimize the microwave extraction technology for total flavonoids from Scutellaria barbata by response surface methodology and to evaluate their in vitro activities.Methods Based on the single-factor test results,Scutellaria barbata were used as raw material,the effects of five main factors:the extraction times,the ethanol concentration,the solid-liquid,the microwave extraction time and the microwave power on the yield of total flavonoids were investigated.The antioxidant activities of the extract were studied by DPPH method and ABTS method,and the acetylcholinesterase inhibitory of the extractwas evaluated by improved Ellman method.Results The optimum extraction process of total flavonoids was as follows:the ethanol concentration was 80%,the microwave power was 525 W,the solid-liquid ratio was 1∶24(mg/m L),the microwave extraction time was 13 min.Under these conditions,the actrual yieid of total flavonoids was 10.89%.The total flavonoids from Scutellaria barbata exhibited a strong antioxidant activity and certain inhibitory activities on acetylcholinesterase.Conclusion This method is reasonable and stable,which can be used for the analysis of total flavonoids from Scutellaria barbata.
