不同比例甘草附子配伍对乌头碱及甘草苷溶出率的影响及镇痛抗炎作用探究 被引量:11

摘要 目的观察不同比例附子甘草配伍组对其主要药效成分乌头碱和甘草苷溶出率影响,并探讨甘草附子配伍后其镇痛、抗炎作用。方法采用紫外-可见分可光度法,分别测绘乌头碱及甘草苷标准曲线。用乌头碱和甘草苷为对照品,分别设置甘草∶附子——1∶0、3∶1、2∶1、1∶1、1∶2、1∶3、0∶1 7个比例组。将小鼠随机分为14组,雌雄各半,每组9只,分别为空白对照组、去痛片阳性对照组、甘草单煎低、中、高剂量组、附子单煎低、中、高剂量组、甘草附子3∶1低、中、高剂量组和甘草附子1∶3低、中、高剂量组。通过热板法和醋酸扭体法测其镇痛效果,二甲苯急性致炎法测定其抗炎效果。结果乌头碱线性回归方程:y=0.028 5x-0.005 7 (R2=0.999 6),0.001~0.008 mg/m L范围内线性关系良好;甘草苷线性回归方程:y=53.146x-0.003 6(R2=0.999 9),0.002~0.001 5 mg/m L范围内线性关系良好。醋酸扭体法镇痛和二甲苯炎症实验中,甘草配伍组1∶3组效果最好。热板法物理镇痛实验中,甘草附子配伍组3∶1效果最好。结论随甘草苷越多,乌头碱溶出量有增大趋势。其中甘草∶附子为3∶1时水煎液乌头碱的溶出量最多,含量为0.002 1%;甘草比附子为1∶3时水煎液甘草苷的溶出量最多,含量为12.50%。同时,甘草苷可能是解痉、止痛抗炎的主要成分;乌头碱是抑制痛觉神经减轻疼痛的主要成分。 Objective To observe the dissolution rate o f aconitine and glycyrrhizin in different proportion of aconite and Radix Glycyrrhizae compatibility group, and to explore the analgesic and anti-inflammatory effects of aconitine and glycyrrhizin after different proportions of compatibility of aconite and Radix glycyrrhizae. Methods The standard curves of aconitine and glycyrrhizin were determined by ultraviolet-visible spectrophotometry. Using aconitine and glycyrrhizin as control, seven proportion groups of liquorice:aconite-1:0, 3:1, 2:1, 1:1, 1:2, 1:3, 0:1 were set up respectively. The mice were randomly divided into 14 groups, half male and half female, with 9 mice in each group. They were blank control group, anti-pain tablet positive control group, liquorice decoction low,medium and high dose group, Aconite Decoction low, medium and high dose group, Radix Aconiti Glycyrrhizae 3:1 low, medium and high dose group and Radix Aconiti Glycyrrhizae 1:3 low, medium and high dose group. The analgesic effect was measured by hot plate method and acetic acid writhing method, and the anti-inflammatory effect was determined by xylene acute inflammation method.Results The linear regression equation of aconitine was y=0.0285 x-0.0057(R2=0.9996), the linear relationship was good in the range of0.001-0.008 mg/ml;the linear regression equation of glycyrrhizin was y=53.146 x-0.0036(R2=0.9999), and the linear relationship was good in the range of 0.002-0.0015 mg/ml. In the experiment of acetic acid writhing analgesia and xylene inflammation, Glycyrrhiza compatibility group 1:3 had the best effect. In the hot-plate physical analgesia experiment, the compatibility group 3:1 of Radix Glycyrrhizae and Radix Aconiti was the best. Conclusion With the increase of glycyrrhizin, the dissolution of aconitine tends to increase. The content of aconitine in water decoction was 0.0021% when the ratio of Radix Aconiti to Radix Aconiti was 3:1, and the content of Glycyrrhizae Glycyrrhizae was 12.50% when the ratio of Radix Aconiti to Radix Aconiti was 1:3. At the same time,glycyrrhizin may be the main component of spasmolysis, pain relief and anti-inflammatory;aconitine is the main component of pain relief by inhibiting pain nerve.
