逍遥散对抑郁模型大鼠下丘脑、结肠中P物质和血管活性肠肽的影响 被引量:18

Effects of Xiaoyaosan on substance P and vasoactive intestinal peptide in hypothalamus and colon of depression model rats

摘要 目的:探讨逍遥散对慢性轻度不可预见性应激(CUMS)模型大鼠下丘脑、结肠中P物质(SP)和血管活性肠肽(VIP)含量的影响。方法:40只SD大鼠随机分为正常组、模型组、逍遥散组、氟西汀组,每组10只。除正常组外,其余3组大鼠均接受CUMS的方式造模,共6周。逍遥散组、氟西汀组在造模同时分别给予逍遥散、氟西汀进行干预治疗。造模结束后次日,取大鼠下丘脑、结肠,采用ELISA法测定SP和VIP的含量。结果:与正常组比较,模型组大鼠下丘脑与结肠SP含量显著升高、VIP含量显著降低(P<0.01);与模型组比较,逍遥散组和氟西汀组下丘脑与结肠SP含量显著降低、VIP含量显著升高(P<0.01,P<0.05)。结论:逍遥散可能是通过调节脑肠肽SP、VIP的含量,达到改善抑郁状态与胃肠功能的作用。 Objective:To investigate the effects of Xiaoyaosan on substance P(SP)and vasoactive intestinal peptide(VIP)contents in colon and hypothalamus of chronic mild unpredictable stress(CUMS)-induced rats.Methods:Forty SD rats were randomly divided into control group,model group,Xiaoyaosan group and fluoxetine group with 10 rats in each group.Except for the control group,rats in the other three groups were subjected to CUMS for 6 weeks.Xiaoyaosan group and fluoxetine group were treated with Xiaoyaosan and fluoxetine,respectively.The contents of SP and VIP in hypothalamus and colon of rats were measured by enzyme linked immunosorbent assay(ELISA).Results:Compared with the control group,the SP content in hypothalamus and colon of model rats increased significantly and the VIP content decreased significantly(P<0.01);after 6 weeks of intervention treatment,compared with the model group,the SP content in hypothalamus and colon of Xiaoyaosan group and fluoxetine group decreased significantly and the VIP content increased significantly(P<0.01,P<0.05).Conclusion:Xiaoyaosan could improve depression and gastrointestinal function by regulating the content of SP and VIP.
