针刀干预对颈椎病兔软骨终板基质金属蛋白酶13、聚集蛋白聚糖、Ⅱ型胶原的影响 被引量:6

Effects of miniscalpel-needle therapy on matrix metalloproteinase 13,aggrecan and typeⅡcollagen in cartilage endplate of rabbit model with cervical spondylosis

摘要 目的:观察针刀干预对颈椎病兔软骨终板基质金属蛋白酶13(MMP-13)、聚集蛋白聚糖(AGG)、Ⅱ型胶原(COL-Ⅱ)的影响,探讨针刀治疗颈椎病可能的疗效机制。方法:将40只新西兰兔随机分为空白组、模型组、电针组、针刀组,每组10只,采用长期异常应力的方法建立颈椎病兔模型,通过X线验证造模是否成功。造模成功后针刀组行针刀干预,对照组为电针组,采用Real-time PCR法和Western Blot法检测软骨终板AGG、COL-Ⅱ、MMP-13的基因和蛋白表达水平。结果:与空白组比较,模型组AGG、COL-ⅡmRNA及蛋白表达显著下降(P<0.01,P<0.05),MMP-13 mRNA及蛋白表达显著上升(P<0.01),电针组COL-ⅡmRNA表达显著下降(P<0.05);与模型组比较,电针组AGG mRNA及蛋白表达显著上升(P<0.05),MMP-13 mRNA及蛋白表达显著下降(P<0.05),针刀组AGG、COL-ⅡmRNA及蛋白表达显著上升(P<0.01,P<0.05),MMP-13 mRNA及蛋白表达显著下降(P<0.05,P<0.01)。结论:针刀治疗颈椎病可以调节AGG、COL-Ⅱ、MMP-13基因和蛋白的表达,修复退变的软骨终板,这可能是针刀治疗颈椎病的效应机制之一。 Objective:To observe the effects of miniscalpel-needle therapy on matrix metalloproteinase 13(MMP-13),aggrecan(AGG)and typeⅡcollagen(COL-Ⅱ)in cartilage endplate of rabbit model of cervical spondylosis,and to explore the possible effect mechanism of miniscalpel-needle in the treatment of cervical spondylosis.Methods:Forty New Zealand rabbits were randomly divided into blank group,model group,electro-acupuncture group and miniscalpel-needle group(10 in each group).The rabbit model of cervical spondylosis was established by long-term abnormal stress method and verified by X-ray.After successful modeling,the miniscalpel-needle group was treated with miniscalpel-needle,and the control group was an electroacupuncture group.The mRNA and protein expression levels of cartilage end plates AGG,COL-Ⅱand MMP-13 were detected by Real-time PCR and Western Blot.Results:Compared with the blank group,the expression of AGG and COL-ⅡmRNA and protein in the model group decreased significantly(P<0.01,P<0.05),while the expression of MMP-13 mRNA and protein increased significantly in the model group(P<0.01);The expression of COL-ⅡmRNA decreased in the electroacupuncture group(P<0.05).Compared with the model group,the expression of AGG mRNA and protein in the electroacupuncture group increased(P<0.05),while the expression of MMP-13 mRNA and protein decreased(P<0.05);In the miniscalpel-needle group,the expression of AGG and COL-ⅡmRNA and protein increased significantly(P<0.01,P<0.05),while the expression of MMP-13 mRNA and protein decreased(P<0.05,P<0.01).Conclusion:Miniscalpel-needle therapy for cervical spondylosis can regulate the expression of AGG,COL-Ⅱ,MMP-13 mRNA and protein,repair the degenerated cartilage endplate.which may be one of the effect mechanisms of miniscalpel-needle therapy for cervical spondylosis.
