摘要
目的:通过观察抗纤益心浓缩丸(KYP)对扩张型心肌病(DCM)大鼠心肌纤维化的影响,探讨KYP抑制心肌纤维化的作用机制。方法:采用自主饮用呋喃唑酮水溶液建立大鼠DCM模型。造模成功后将大鼠随机分为模型组,KYP高、中、低剂量组,卡托普利组,另设有正常组,每组8只,各组给予相应药物灌胃。干预8周后检测各组大鼠心肌组织Masson染色及心肌胶原容积分数(CVF);免疫组化检测心肌组织Ⅰ、Ⅲ型胶原蛋白的表达;Westen Blot检测α-平滑肌肌动蛋白(α-SMA)、结缔组织生长因子(CTGF)蛋白表达。结果:与模型组比较,KYP高、中、低剂量组及卡托普利组可明显降低CVF及Ⅰ、Ⅲ型胶原表达(P<0.01,P<0.05),改善心肌纤维化病理,KYP高、中剂量组及卡托普利组可显著抑制α-SMA、CTGF蛋白表达水平(P<0.01,P<0.05)。结论:KYP可抑制Ⅰ、Ⅲ型胶原增生,可能通过抑制心肌成纤维细胞转化及CTGF表达。
Objective:To investigate the effects of concentrated Kangxian Yixin Pills(KYP)on myocardial fibrosis in rats by observing the effect of concentrated KYP on myocardial fibrosis in rats with dilated cardiomyopathy(DCM).Methods:Rat DCM model was established by autonomous drinking furazolidone aqueous solution.After successful modeling,the rats were randomly divided into model group,KYP high,medium and low dose groups,captopril group,and control group,8 rats in each group.Each group was given the corresponding drugs.After 8 weeks of intervention,masson staining and myocardial collagen volume fraction(CVF)were detected in the myocardial tissue of each group.Immunohistochemistry was used to detect the expression of typeⅠandⅢcollagen in myocardial tissue.Western Blot was used to detect the expression ofα-SMA and CTGF protein.Results:Compared with the model group,KYP high,medium and low dose groups and captopril group could significantly reduce the expression of CVF,typeⅠandⅢcollagen(P<0.01,P<0.05),improve the pathology of myocardial fibrosis,KYP high and medium dose groups and captopril group inhibit the expression ofα-SMA and CTGF protein(P<0.01,P<0.05).Conclusion:KYP can inhibit the proliferation of typeⅠandⅢcollagen,which may be related to the inhibition of myocardial fibroblast transformation and CTGF expression.
