摘要
目的:建立赤茯苓HPLC指纹图谱,并进行相似度评价,结合指纹图谱对15批赤茯苓样品进行主成分分析和聚类分析,从而进行整体评价。为赤茯苓质量评价提供理论基础。方法:采用Sharpsil-UC18色谱柱(250mm×4.6mm,5μm);流动相:乙腈-0.2%冰醋酸梯度洗脱;流速1.0mL/min,检测波长245nm,柱温30℃。结果:建立了具有21个共有峰的赤茯苓HPLC指纹图谱,获得了评价其质量的大量数据,以15批赤茯苓指纹图谱的共有模式作为赤茯苓对照指纹图谱,15批赤茯苓的相似度值为0.795~0.991,存在明显差异。通过聚类分析和主成分分析方法,能明显地将云南产地赤茯苓与地理距离差异大的其他产地赤茯苓区分开来。结论:所建立的HLPC指纹图谱方法结合化学计量学可以用于不同产地赤茯苓的质量评价。
Objective:To establish the HPLC fingerprint of Rubra Poria cocos and evaluate its similarity.The principal component analysis(PCA)and cluster analysis(HCA)were used to evaluate 15 batches of red Poria cocos.Provide the oretical basis for the quality evaluation of Rubra Poria cocos.Methods:The chromatographic column was Sharpsil-U C18(250 mm×4.6 mm,5μm),the flow phase was acetonitrile(A)-0.2%Acetic acid glacial solution(B),the flow rate was 1.0 mL/min,the detection wavelength was 245 nm,and the column temperature was 30℃.Results:The HPLC fingerprint of Rubra Poria cocos containing 21 common peaks was established and important digital information was obtained that could be used to distinguish the quality.Using common pattern of 15 batches of Rubra Poria cocos as the standard fingerprint,the similarity value of 15 batches of Rubra Poria cocos were 0.795~0.991,which was significantly different.Through cluster analysis and principal component analysis,it can distinguish the Rubra Poria cocos of Yunnan Province from those of other places with great difference in geographical distance.Conclusion:The established HLPC fingerprint method combined with chemometrics can be used for the quality evaluation of Rubra Poria cocos from different habitats.
