冬凌草提取物治疗非酒精性脂肪性肝病的作用及机制分析 被引量:2

摘要 目的基于线粒体功能探讨冬凌草对非酒精性脂肪性肝病(NAFLD)的干预效果及相关作用机制.方法60只SD健康雄性大鼠,随机选取10只作为正常组,其余50只建立非酒精性脂肪性肝病模型,随机分为模型组、药物对照组、低剂量组、中剂量组、高剂量组.正常组、模型组大鼠使用生理盐水进行干预,药物对照组大鼠使用非诺贝特溶液进行干预,低剂量组、中剂量组、高剂量组大鼠使用不同剂量冬凌草提取物溶液进行干预.检测甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL)、低密度脂蛋白胆固醇(LDL)、游离脂肪酸(FFA)、丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、丙转氨酶(ALT)、谷草转氨酶(AST)水平;检测肝脏组织细胞凋亡情况,Bcl-2、Bax、Caspase3相对表达量.结果药物对照组、高剂量组大鼠血清、肝脏中TG、TC、FFA、LDL、ALT、AST、MDA水平均低于低剂量组、中剂量组,HDL、SOD、GSH-Px水平均高于低剂量组、中剂量组(P<0.05).药物对照组、高剂量组大鼠肝组织细胞凋亡率低于低、中剂量组(P<0.05).药物对照组、高剂量组大鼠Bcl-2相对表达量高于低、中剂量组,Bax、Caspase3相对表达量低于低、中剂量组(P<0.05).结论冬凌草提取物对非酒精性脂肪性肝病模型大鼠进行干预,能够改善大鼠血清、肝脏中脂质含量及肝功能,减轻大鼠肝组织细胞氧化应激损伤,并能调控线粒体凋亡通路蛋白Bcl-2、Bax、Caspase3的表达,抑制大鼠肝组织细胞凋亡,延缓非酒精性脂肪性肝病的发生发展. Objective To explore the intervention effect and mechanism of rabdosia rubescens on nonalcoholic fatty liver based on mitochondrial function.Methods 60 SD healthy male rats,10 of which were randomly selected as normal group-The other 50 rats were randomly divided into model group,drug control group.low dose group,middle dose group and high dose group.Rats in normal group and model group were intervened with normal saline,rats in drug control group were intervened with fenofibrate solution,rats in low dose group,middle dose group and high dose group were intervened with different doses of rabdosia mbcsccns extract solution.The level of triglyceride(TG)was detected by lipase method.Total cholesterol(TC)level was determined by cholesterol oxidase assay.The levels of high density lipoprotein cholesterol(HPL),low density lipoprotein cholesterol(LPL),free fatty acid(FFA),malondialdehyde(MPA),superoxide dismutase(SOP)and glutathione peroxidase(GSH-Px)were measured by enzyme-linked immunosorbent assay.ALT and AST levels were detected by turbidimetry.TUNEL method was used to detect the apoptosis of liver cells,and Western blot was used to detect the relative expressions of bcl-2,Bax and caspase3.Results The levels of TG,TC,FFA,LPL,ALT,ASF and MPA m serum and liver of drug control group and high dose group were lower than those of low dose group and middle dose group,ansi the levels of HDL,SOP and GSM PX were higher than those oflow dose group and middle dose group(P<0.05).The apoptosis rate of hcpatocytcs in the drug control group and high dose group were lower than that in the low and middle dose group(P<0.05).The relative expression of Bel-2 in the drug control group and high dose group were higher than that in the low and middle dose groups,and the relative expression of Bax and caspasc-3 was lower than that in the low and middle dose groups(P<0.05).C onclusion By using rabdosia rubescens extract to intervene the model rats of nonalcoholic fatty liver disease,it can improve the lipid content and liver function in serum and liver,reduce the oxidative stress injury of liver tissue cells,regulate the expression of Bcl-2,Bax and caspase 3.inhibit the apoptosis of liver tissue cells and delay the development of nonalcoholic fatty liver disease.
