妊娠期糖尿病患者外周血中Th1/Th2细胞及相关细胞因子表达水平变化的研究

Changes of Th1/Th2 Cells and Related Cytokines Expression in Peripheral Blood of Gestational Diabetes Mellitus

目的:观察妊娠期糖尿病患者外周血中Th1/Th2细胞及相关细胞因子表达水平的变化。方法:选择在笔者所在医院就诊的50例妊娠期糖尿病患者为观察组,50例健康妊娠的孕妇为对照组,收集观察组和对照组产妇的外周血,通过流式细胞术分析Th1/Th2细胞比例,采用ELISA检测白细胞介素-2(IL-2)、白细胞介素-4(IL-4)、白细胞介素-10(IL-10)、干扰素-γ(IFN-γ)的表达水平。结果:观察组患者外周血中Th1/Th2细胞的比例高于对照组,差异有统计学意义(P<0.05);观察组患者外周血中IL-2、IFN-γ的表达水平高于对照组,IL-4、IL-10的表达水平低于对照组,差异均有统计学意义(P<0.05)。结论:妊娠期糖尿病患者外周血中Th1/Th2细胞比例失衡,Th1细胞因子表达升高,Th2细胞因子表达降低,Th1/Th2免疫平衡的改变可能是妊娠期糖尿病免疫诊断和干预的新靶点,为妊娠期糖尿病的治疗提供试验依据。

Objective:To observe the expression of Th1/Th2 cells and related cytokines in the peripheral blood of patients with gestational diabetes mellitus.Method:Totally 50 cases of gestational diabetes in our hospital were selected as the observation group and 50 cases of healthy pregnant women as the control group.The peripheral blood of the observation group and the control group were collected.The proportion of Th1/Th2 cells was analyzed by flow cytometry.The levels of IL-2,IL-4,IL-10,IFN-γwere detected by ELISA The level of expression.Result:The proportion of Th1/Th2 cells in the peripheral blood of the observation group was higher than that of the control group,the difference was statistically significant(P<0.05).The expression level of IL-2 and IFN-γin the peripheral blood of the observation group were higher than those of the control group,and the expression level of IL-4 and IL-10 were lower than those of the control group,the differences were statistically significant(P<0.05).Conclusion:The imbalance of Th1/Th2 cell proportion,the increase of Th1 cytokine expression and the decrease of Th2 cytokine expression in the peripheral blood of patients with gestational diabetes mellitus may be the new target of immunodiagnosis and intervention of gestational diabetes mellitus,and provide experimental basis for the treatment of gestational diabetes mellitus.
