摘要
为了探讨miRNA-499、miRNA-146、miRNA-149基因多态性与结直肠癌远处转移的相关性,本研究收集了376例结直肠癌患者,分为无远处转移组(n=167)和有远处转移组(n=180),同时搜集健康对照组患者(n=193)。采用PCR-限制性内切酶片段长度多态性技术对患者外周血miR-146a rs2910164 G>C、miR-149rs2292832 C>T和miR-499 rs3746444 T>C进行基因型分析。结果证实携带miR-146a rs2910164 CC基因型的结直肠癌患者在远处转移组和健康对照组(CC vs CG,p=0.006;CC vs GG,p=0.047)以及无远处转移组和远处转移组(CC vs CG,p=0.02;CC vs GG,p=0.012)之间的差异均具有统计学意义。病理分期分析显示携带miR-146a rs2910164 CC基因型的结直肠癌患者在病理低分化组和高分化组(CC vs CG,p=0.03;CC vs GG,p=0.047)之间差异具有统计学意义。因此本研究推测携带miR-146a rs2910164 CC基因型的结直肠癌患者分化程度更低,更容易发生远处转移。
The purpose of this study was to investigate the correlation between miRNA-499,miRNA-146,miRNA-149 gene polymorphism and distant metastasis of colorectal cancer.We collected 376 patients with colorectal cancer,divided the patients into without distant metastases group(n=167),and distant metastasis group(n=180),while collecting healthy physical examination as a healthy control group(n=193).Mir-146 a rs2910164 G>C,mir-149 rs2292832 C>T and mir-499 rs3746444 T>C in peripheral blood of patients were analyzed using pcr-restriction enzyme digestion fragment length polymorphism(RFLP).Study showed colorectal cancer patients with mir-146 a rs2910164 CC genotype were both in the distant metastatic group and the healthy control group(CC vs CG,p=0.006;CC vs GG,p=0.047)and without distant metastatic group and the distant metastatic group(CC vs CG,p=0.02;CC vs GG,p=0.012).The difference between those were statistically significant.All patients were statistically analyzed by tumor staging.The difference between the pathological hypodifferentiation group and hyperdifferentiation group(CC vs CG,p=0.03;CC vs GG,p=0.047)was statistically significant.We can speculated that colorectal cancer patients with mir-146 a rs2910164 CC genotype had lower differentiation degree and were more prone to distant metastasis.
