脾切除小鼠肝脏和外周血免疫细胞的动态变化 被引量:3

摘要 目的探讨脾切除小鼠肝脏和外周血免疫细胞的动态变化。方法32只雄性C57BL/6小鼠分为假手术组、脾切除1周组、脾切除2周组、脾切除4周组,每组8只。于术后第1周、2周、4周检测小鼠肝脏质量和肝指数,用流式细胞术检测外周血和肝脏免疫细胞(CD3-CD19+B细胞、CD3+CD4+T细胞、CD3+CD8+T细胞、CD11b+Ly6C hi单核细胞、CD11b+Ly6C med单核细胞、CD11b+Ly6G+粒细胞、CD11c+MHCⅡ+DC细胞、CD11b int F4/80 hi枯否细胞、CD11b hi F4/80 int巨噬细胞、CD11b hi F4/80 int Ly6C hi巨噬细胞、CD11b hi F4/80 int Ly6C low巨噬细胞)的比例。结果与假手术组相比,脾切除1周、2周、4周组小鼠肝脏质量与肝指数均无显著差异(P>0.05);脾切除1周组肝脏CD11b+Ly6G+粒细胞显著升高(t=-2.339,P<0.01);脾切除4周组肝脏CD3+CD8+T细胞显著升高(t=-5.092,P<0.01);脾切除1周、2周、4周组肝脏CD3+CD4+/CD3+CD8+均显著降低(t=2.785,2.45,-3.549,均P<0.05);脾切除1周组外周血CD11b+Ly6C hi单核细胞(t=-3.321,P<0.05)、CD11b+Ly6G+粒细胞(t=-2.845,P<0.05)显著升高;其余细胞均无显著变化。结论脾切除术后肝脏和外周血免疫细胞比例发生变化,提示脾脏在维持肝脏区域免疫稳态中具有重要作用。 Objective To investigate the dynamic changes of immune cells in liver and peripheral blood of mice after splenectomy.Methods Thirty-two male C57BL/6 mice were divided into four groups:sham operation group(n=8),one week after splenectomy group(n=8),two weeks after splenectomy group(n=8)and four weeks after splenectomy group(n=8).At week 1,2,4 after operation,the liver weight and the liver index of mice were measured.The immune cells CD3-CD19+B cells,CD3+CD4+T cells,CD3+CD8+T cells,CD11b+Ly6C hi monocytes,CD11b+Ly6C med monocytes,CD11b+Ly6G+granulocytes,CD11c+MHCⅡ+DC,CD11b int F4/80 hi Kupffer cells,CD11b hi F4/80 int macrophages,CD11b hi F4/80 int Ly6C hi macrophages,CD11b hi F4/80 int Ly6C low macrophages in peripheral blood and liver were detected by flow cytometry.Results There were no significant differences in liver weight and liver index between sham operation group and splenectomy groups(P>0.05).The percentages of CD11b+Ly6G+granulocytes(t=-2.339,P<0.01)in liver in one week after splenectomy group and the percentages CD3+CD8+T cells(t=-5.092,P<0.01)in liver in four weeks after splenectomy group were both significantly higher than those in sham operation group.However,the ratio of CD3+CD4+T cells and CD3+CD8+T cells in liver was significantly decreased in one,two,and four weeks after splenectomy groups compared with sham operation group(t=2.785,2.45,-3.549,all P<0.05).The percentages of CD11b+Ly6C hi monocytes(t=-3.321,P<0.05)and CD11b+Ly6G+granulocytes(t=-2.845,P<0.05)in peripheral blood in one week after splenectomy group were significantly higher than those in sham operation group.There was no significant change in other cells between sham operation group and splenectomy groups.Conclusion The ratio of immune cells in the liver and the peripheral blood changes after splenectomy,which suggests that the spleen plays an important role in maintaining the immune homeostasis in the liver.
