蒲公英甾醇通过TGF-β/Smad通路对小鼠痤疮模型炎症因子水平、胸腺组织形态的影响 被引量:15

Effects of taraxasterol on inflammatory factor levels and morphology of thymus in acne model of mice through TGF-β/Smad pathway

摘要 目的探讨蒲公英甾醇是否通过转化生长因子β(transforming growth factor-β,TGF-β)/Smad)通路对小鼠痤疮模型炎性因子水平、胸腺组织形态的影响。方法将C57BL/6小鼠分为对照组(A组)、模型组(B组)、蒲公英甾醇低剂量组(C组)、蒲公英甾醇高剂量组(D组);观察各组小鼠耳部变化;苏木精-伊红染色法(hematoxylin-eosin staining,HE)观察各组小鼠耳组织和胸腺组织病理变化;酶联免疫吸附法(enzyme linked immunosorbent assay,ELISA)检测各组小鼠血清中白细胞介素1β(interleukin-1β,IL-1β)和IL-8表达水平;免疫组织化学检测各组小鼠耳组织中肿瘤坏死因子α(tumor necrosis factor-α,TNF-α)阳性细胞表达率;Western blot检测各组小鼠耳组织中转化生长因子β1(transforming growth factor-β1,TGF-β1)、Smad3蛋白表达。结果与B组相比,C组和D组减轻了痤疮小鼠耳部肿胀、发红和红斑、质地干燥粗糙等状态,且耳朵厚度也明显降低(P<0.05);蒲公英甾醇经灌胃治疗后,痤疮小鼠的炎症得到了有效缓解,并且浸润的炎症细胞数量明显减少,表皮增厚也减轻;与B组相比,C组和D组增加了小鼠痤疮模型胸腺皮质厚度和淋巴细胞数量(均P<0.05);蒲公英甾醇明显降低了小鼠血清中IL-1β和IL-8的表达水平(P<0.05或P<0.01),且小鼠耳组织中TNF-α的阳性表达率减少(P<0.05);与B组相比,C组和D组使小鼠耳组织中TGF-β1、Smad3蛋白表达下调(均P<0.05)。结论蒲公英甾醇对小鼠耳廓痤疮模型有较好的治疗效果,调节炎性因子的表达、改善小鼠胸腺组织形态,其可能与TGF-β/Smad通路有关。 Objective To investigate the effect of taraxasterol on the level of inflammatory factors and morphology of thymus in acne model mice through transforming growth factor-β(TGF-β)/Smad pathway.Methods C57BL/6 mice were divided into:control group(group A),model group(group B),taraxasterol low-dose group(group C),taraxasterol high-dose group(group D).The ear changes of mice in each group were observed;hematoxylin-eosin staining(HE)staining was used to observe the pathological changes of ear tissue and thymus tissue of mice in each group.Enzyme linked immunosorbent assay(ELISA)was used to detect the expression levels of interleukin-1β(IL-1β)and IL-8 in the serum of mice in each group.Immunohistochemical was used to detect of the expression rate of tumor necrosis factor-α(TNF-α)positive cells in the ear tissues of mice in each group.Western blot was used to detect the expression of transforming growth factor-β1(TGF-β1)and Smad3 protein in the ear tissue of mice in each group.Results Compared with group B,group C and group D reduced the ear swelling,redness and erythema,dry and rough texture of acne mice,and the ear thickness was significantly reduced(P<0.05).After taraxasterol treatment by gavage,the inflammation in acne mice was effectively alleviated,and the number of infiltrating inflammatory cells was significantly reduced,and the thickening of the epidermis was also reduced.Compared with group B group,group C and group D increased thymic cortical thickness mice acne and lymphocytes(both P<0.05);Taraxasterol significantly reduced the level of expression of IL-1βand IL-8 of mouse serum(P<0.05 or P<0.01),and the ear tissues of mice TNF-αexpression is reduced(P<0.05).Compared with group B,group C and group D down-regulated the expression of TGF-β1 and Smad3 protein in mouse ear tissues(both P<0.05).Conclusion Taraxasterol had a good therapeutic effect on mouse auricular acne model,regulates the expression of inflammatory factors and improves the morphology of mouse thymus,which may be related to TGF-β/Smad pathway.
