摘要
目的研究1株南海来源真菌Penicillium chrysogenum的次级代谢产物及其抗菌活性。方法运用硅胶柱层析、Sephadex LH-20凝胶柱层析和HPLC半制备等方法对次级代谢产物进行分离和纯化;通过NMR、MS等方法鉴定化合物结构;利用抗菌活性模型对其进行活性评价。结果从真菌P.chrysogenum中分离鉴定了5个化合物,包括1个生物碱taichunamide H(1),1个带有脂肪链的蒽醌1-O-methyl-averantin(2),2个聚酮化合物versicones A–B(3~4)以及1个联苯醚diorcinol(5);其中,2对致病菌Canidia albicans和Staphylococcus aureus具有较强的抑制活性,MIC值分别为6.25和12.5μmol·L^-1;5对海洋污损菌Photobacterium halotolerans显示抑制活性,MIC值为50μmol·L^-1。结论化合物2显示较强的抗菌活性,具有潜在的开发研究价值,且化合物3和4为首次从青霉属真菌中分离得到。
Objective To study the chemical structures and antimicrobial activities of the marine-derived fungus Penicillium chrysogenum collected from the South China Sea.Methods The compounds were separated and purified by silica gel column chromatography,Sephadex LH-20 gel,and semi-HPLC.Their structures were identified by modern spectroscopic analysis including NMR and MS methods.In addition,the antimicrobial activities of the isolated compounds were also evaluated.Results Five compounds were isolated and identified from a marine-derived fungus P.chrysogenum,including one alkaloid with complex structure,taichunamide H(1),one anthraquinone containing aliphatic chain,1-O-methyl-averantin(2),two xanthonoids,versicones A-B(3~4),and one diphenyl ether,diorcinol(5).Compound 2 showed significant antimicrobial activities against pathogenic bacteria Canidia albicans and Staphylococcus aureus with MIC values of 6.25 and12.5μmol·L^-1,respectively.Compound 5 displayed inhibitory activity against stained bacteria Photobacterium halotolerans with an MIC value of 50μmol·L^-1.Conclusion Compound 2 showed strong antimicrobial activity with potential development and research value,and compounds 3 and 4 were isolated from fungus Penicillium sp.for the first time.
