褐藻糖胶与海洋深层水联合用药对Ⅱ型糖尿病大鼠认知障碍的干预作用

Combination of fucoidan and deep sea water ameliorates type Ⅱdiabetes and associated-cognitive impairment

摘要 目的以Ⅱ型糖尿病(Type 2 diabetes mellitus,T2DM)大鼠为模型,研究褐藻糖胶(fucoidan,FPS)与海洋深层水(deep sea water,DSW)联合用药对胰岛素抵抗及认知障碍的干预作用。方法采用高糖高脂饮食联合链脲佐菌素(streptozotocin,STZ)诱导T2DM大鼠模型,然后给予FPS、DSW或DSW与FPS 3个剂量联用,给药期间监测各组大鼠体质量以及摄食摄水等一般情况,给药4周后,进行Morris水迷宫实验。实验结束后取血清,测定空腹血糖(fasting blood glucose,FBG)、空腹胰岛素(fasting insulin,FINS)、血脂四项(总胆固醇(total cholesterol,TC)、甘油三酯(triglyceride,TG)、低密度脂蛋白胆固醇(low-density lipoproteincholesterol,LDL-C)、高密度脂蛋白胆固醇(high-density lipoprotein-cholesterol,HDL-C))等指标,计算胰岛素抵抗指数(Homeostasis model assessment insulin resistance,HOMA-IR)。结果与空白组比较,模型组大鼠的FBG、TC、TG、LDL-C、FINS、HOMA-IR均有统计学差异(P<0.05或P<0.01),Morris水迷宫实验中的逃避潜伏期明显延长,在目标象限的停留时间显著减少。而与模型组比较,DSW、FPS及DSW与FPS联用治疗均可以明显改善大鼠FBG、TC、TG、HDL-C、LDL-C、FINS、HOMA-IR等指标,显著缩短逃避潜伏期,提高目标象限的停留时间,且DSW与FPS高剂量联用组的改善作用更显著。结论FPS与DSW联用可以更有效地缓解胰岛素抵抗,且在改善T2DM相关的认知障碍方面具有较好疗效。 Objective To study the intervention effect of combination of fucoidan(FPS)and deep sea water(DSW)on insulin resistance and cognitive dysfunction using T2 DM rats as a model.Methods The T2 DM model rats were induced by high glucose and fat diet combined with streptozotocin(STZ),then the rats were administrated with FPS,DSW or combination of FPS and DSW.During the administration,the body weight and water intake in each group were monitored.The Morris Water Maze experiment was performed after four weeks of administration.At the end of the experiment,the rats were anesthetized and the serum was got to detect the levels of fasting blood glucose(FBG),fasting insulin(FINS),serum lipids(total cholesterol(TC),triglyceride(TG),low-density lipoprotein cholesterol(LDL-C),and high-density lipoprotein cholesterol(HDL-C)).Results Compared with the control group,the levels of FBG,TC,TG,LDL-C,FINS and HOMAIR of rats in model group were statistically significant(P<0.05 or P<0.01),the escape latency in Morris water maze experiment was significantly prolonged,the residence time in the target quadrant was significantly reduced.Compared with model group,DSW,FPS and DSW combined with FPS could significantly improve the indicators of FBG,TC,TG,HDL-C,LDL-C,FINS,HOMA-IR in rats,significantly shorten the escape latency and increased the residence time of the target quadrant.What’s more,co-treatment with DSW and FPS had a greater improving effect than DSW or FPS alone.Conclusion The combination of FPS and DSW could more effectively alleviate insulin resistance in T2 DM rats,and has a better effect on modulation of the cognitive impairment.
