摘要
目的探究丹参乙酸镁(magresiumn lthospermutle B.MLB)平衡ThI.Th2细跑因子治疗系膜增生性肾炎大鼠肾功能受损及肾组织损伤影响。方法选取100只雄性SD大鼠,体重(200+30)g.随机选取20只作为正常对照组(Control),80只制成系膜增生性肾小球肾炎模型,并分为模型组(MsPGN).低剂量丹参乙酸镁组(MIL.B 7.5 mg/kg).中剂量丹参乙酸镁组(MIL.B 15 my/kg)和高剂量丹参乙酸镁组(MILB 30 mg/kg)。检测各组大鼠尿液中24 h蛋白尿、血清肌酐水平、尿素氮的水平;使用HE染色观察组织病理损伤情况;使用RT-PCR检测含半胱氨酸的天冬氨酸蛋白水解酶3(Caspase-3)的表达情况;使用试剂盒检测丙二醛(MDA).超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GPX)水平;使用酶联免疫法(Elisa)检测诱导型一氧化氮合成酶(iN0S).肿瘤坏死因子-a(TNF-a).白介素4(IL-4).白介素-10(IL-10)的表达:使用蛋白质印迹法检测Caspse-3,转化生长因子β(TGF-B)、细胞信号转导分子(Smad2)和磷酸化细胞信号转导分子(p-Smad2)表达。结果相比空白对照组,模型组大鼠24 h蛋白尿,血清肌酸酐、尿素氮,Caspuse-3基因及蛋白表达,MIDA、iNos.TNF-a、TGF-B.p-Smd2/Smad2水平显著升高,SOD.GPX、IL4.IL-10水平显著降低(P<0.05);相比模型组,使用丹参乙酸镁处理各组大鼠24h蛋白尿、血清肌酸酐、尿素氮,Caspuse-3基因及蛋白表达、MDA.iNos.TNF-a.TGF-B、p Smad2/smad2水平显著降低,sOD.GPX.L4.IL-10水平显著升高(P<0.05),且呈浓度依赖性。结论丹参乙酸镁可以通过降低Thi因子升高TH2因子水平,实现降低炎症反应及凋亡因子水平缓解肾组织损伤。
Objective To investigate how magnesium lithospermate B(MLB)balances Thl'Th2 eytokines in the treatment of mesangial proliferative nephritis in rats with impaired renal function and renal tissues damage,Methods A total of 100 male SD rats weighing(200±30)g were ex-amined.20 rats were randomly selected as normal control group(Control),and 80 were made into a model of mesangial proliferative nephritis.And they were divided into a model group(MsPGN),a low-dose MILB group(MLB 7.5 mg/kg),a medium-dose MILB group(MLB 15 mg/kg)and a high-dose MLB group(MLB 30 mg/kg).The levels of 24-h proteinuria,serum creatinine and urea nitrogen in urine of each group were detected.Histopathological damage was observed by HE stai-ning.The expression of cysteine-containing aspartate-speeific protease8-3(Caspase-3)was dected by RT-PCR.The levels of malondialdehyde(MDA),superoxide dismutase(SOD)and glutathi-one peroxidase(GPX)were detected by using test kits.The euzyme-linked immunosorbent assay(ELISA)was performed to deteet the expression of inducible nitrie oxide synthase(iNOS),tumor necrosis factor-a(TNF-a),interleukin4(IL4)and interleukin-10(IL-10).Westerm blotting was conducted to detect the expression of Caspuse-3,transforming growth factor-β(TGF-β),sup-pressor of mothers against deeapentaplegie 2(Smad2)and phosphorylated Smad2(p-Smad2).Results Compared with blank control group,24-h proteinuria,serum creatinine,urea nitrogen,Caspase-3 gene and protein expression,levels of MDA,iNOS,TNF-x,TGF-βand p-Smad2/Smad2 were significantly inereased in model group,while levels of S0D,GPX,IL.4 and IL-10 were signifieantly decreased(P<0.05).Compared with model group,24-h proteinuria,serum creatinine,urea mitrogen,Caspase-3 gene and protein expression,levels of MDA,iNos,TNF-a,TCF-βand p-Smad2/Smad2 were significantly decreased in the groups treated by MLB,while levels of SOD,CPX,L.4 and IL-10 were significantly increased(P<0.05),showing a concen-tration-dependent pattern.Conclusion MLB can reduce inflammatory reaction and apoptotic factors levels and alleviate renal issue damage by lowering Thl factor and inereasing TH2 level.
