miRNA-223与IgE在小鼠鼻腔息肉组织中的差异性表达与意义 被引量:2

摘要 目的探索miRNA-223与IgE在小鼠鼻腔息肉组织中的差异性表达与意义。方法 qPCR检测miR-223和IgE的差异表达;食物藏球实验检测的嗅觉能力;HE染色检测鼻粘膜组织的病理变化;免疫组织化学检测鼻粘膜组织中MPO和OMP的表达;ELISA检测鼻腔灌洗液中细胞因子的表达;Western blot检测鼻粘膜组织中IgE蛋白的表达。结果 miR-223和IgE在鼻息肉患者和鼻息肉小鼠鼻粘膜组织中表达上调(均P<0.001),miR-223inhibitor能抑制miR-223和IgE的表达(均P<0.05);与control组相比,LPS组出现明显的嗅觉障碍、鼻腔中有息肉形成以及MPO+细胞增加和OMP+细胞减少,鼻腔灌洗液中IgE、TNF-α、IL-6的含量增加和鼻粘膜组织中IgE蛋白表达上调(均P<0.01);与LPS组相比,miR-223inhibitor组小鼠的嗅觉障碍缓解、鼻腔粘膜息肉数量减少以及MPO+细胞减少和OMP+细胞增加,鼻腔灌洗液中IgE、TNF-α、IL-6的含量较少和鼻粘膜组织中IgE蛋白表达下调(均P<0.05)。结论 miR-223和IgE在鼻息肉中高表达,且低表达miR-223能显著降低IgE的表达。 Objective to explore the differential expression and significance of mirna-223 and IgE in mice nasal polyps.Methods qPCR was used to detect the differential expression of mir-223 and IgE,olfactory ability of food storage ball test,he staining to detect the pathological changes of nasal mucosa,immunohistochemistry to detect the expression of MPO and OMP in nasal mucosa,ELISA to detect the expression of cytokines in nasal lavage fluid,Western blot to detect the expression of IgE protein in nasal mucosa.Results the expression of mir-223 and IgE was up-regulated in patients with nasal polyps and mice nasal mucosa(P<0.001),and mir-223 inhibitor inhibited the expression of mir-223 and IgE(P<0.05);Compared with control group,LPS group showed significant olfactory disturbance,polyp formation in nasal cavity,increase of MPO+cells and decrease of OMP+cells,increase of IgE,TNF-α,IL-6 in nasal lavage fluid and up-regulation of IgE protein expression in nasal mucosa(P<0.01);Compared with LPS group,mice in mir-223 inhibitor group had less olfactory disturbance,less polyps in nasal mucosa,less MPO+cells and more OMP+cells,less IgE,TNF-α,IL-6 in nasal lavage fluid and down-regulation of IgE protein expression in nasal mucosa(P<0.05).Conclusion mir-223 and IgE are highly expressed in nasal polyps,and low expression of mir-223 can significantly reduce the expression of IgE.
