miRNA-21调控HER-2阳性乳腺癌的血管生成 被引量:3

摘要 目的探讨mi RNA-21 (mi R-21)对HER-2阳性乳腺癌生物学特性以及血管生成的影响。方法收集HER-2阳性乳腺癌标本,通过PCR分析mi R-21的表达,通过CCK8和Transwell实验验证增殖和侵袭能力,通过Western blotting实验验证VEGF表达变化。结果 mi R-21在HER-2阳性乳腺癌中过表达(P<0.001),但其过表达不会影响预后(在所有乳腺癌患者中P=0.47,在HER-2阳性乳腺癌患者中P=0.26)。抑制mi R-21表达可抑制HER-2阳性乳腺癌的增殖(P<0.01)和侵袭(P<0.001),并且抑制mi R-21的表达可抑制血管生成因子VEGF的表达。结论癌基因mi R-21在调控HER-2阳性乳腺癌生物学行为以及血管生成中发挥一定作用。 Objective To study the effects of mir NA-21(Mir-21) on the biological characteristics and whether mi RNA-21 can regulate angiogenesis in HER-2 positive breast cancer. Methods 40 breast cancer tissues were collected. The expression of mir-21 was detected by real-time PCR. Ability of proliferation was carried out by CCK8 assay and the ability of invasion was studied by transwell invasion assay. The expression of VEGF was researched by western blotting assay. Results Mir-21 showed higher expression in breast cancer tissue than in the adjacent noncancer tissue(P<0.001). Overexpression of mi R-21 will not influence the prognosis(P = 0.47 for all breast cancer patients and P = 0.26 for HER-2 positive breast cancer patients). Inhibiting the expression of mi R-21 inhibited the proliferation(P<0.01) and invasion(P<0.001) of HER-2 positive breast cancer cell, it also inhibited the expression of VEGF. Conclusion The oncogene mi R-21 plays significant role in HER-2 positive breast cancer biological behaviour and angiogenesis.
