Establishment of Tissue Culture and Rapid Propagation System for Chenopodium quinoa Stems

摘要 以藜麦带腋芽茎段为外植体,通过外植体消毒、腋芽诱导、不定芽增殖、生根培养和驯化移栽等过程建立藜麦离体再生体系。结果表明,带腋芽茎段的最佳消毒方案为75%酒精30 s、0.1%HgCl29 min;腋芽诱导最佳培养基为MS+6-BA 1.5 mg/L+NAA 0.5 mg/L,该培养基条件下,萌芽率为90.0%,芽体饱满;不定芽增殖最适培养基是MS+6-BA 1.0 mg/L+NAA 0.2 mg/L,该培养基条件下,增殖系数为4.9;最优生根培养基为1/2 MS+IBA 0.5 mg/L,该培养基条件下生根率为76.7%,不定根数量多,根较粗壮;最佳移栽基质为蛭石∶草炭土体积比3∶1,该基质条件下,组培苗移栽成活率可达93.3%、生长高度为2.86 cm。 In this study,the stem segments with axillary buds of Chenopodium quinoa were used as explants,the regeneration system of quinoa in vitro was established by explant disinfection,axillary bud induction,adventitious bud proliferation,rooting culture and domestication transplanting,and so on.The results showed that the best disinfection scheme of stem segment with axillary bud was 75%alcohol for 30 s and 0.1%HgCl2 for 9 min.The best induction medium was MS+6-BA 1.5 mg/L+NAA 0.5 mg/L,and under the condition of this medium,the induction rate was 90%and the bud was plump.The optimum multiplication medium for adventitious bud was MS+6-BA 1.0 mg/L+NAA 0.2 mg/L,and under the condition of this medium,a proliferation coefficient was 4.9.The best rooting medium was 1/2 MS+IBA 0.5 mg/L,and under the condition of this medium,rooting rate reached 76.7%and root number was numerous and stronger.The best transplanting substrate was vermiculite∶peat soil volume ratio 3∶1,and under the condition of this substrate,the survival rate of transplanting tissue culture seedlings could be up to 93.3%and the seedling height was 2.86 cm.
