四妙勇安汤对动脉粥样硬化模型兔自噬相关因子的影响 被引量:6

摘要 目的:考察四妙勇安汤对新西兰动脉粥样硬化家兔血脂生化指标及自噬相关因子Atg5、Beclin1、mTOR mRNA表达的影响,初步阐明其抗AS的作用机制。方法:将雄性新西兰兔随机分为正常对照组、模型组、四妙勇安汤小、中、大剂量组和辛伐他汀组,每组6只。除正常对照组外,其他各组均建立AS模型,造模后连续给药6周,检测血清中TG、TC、LDL-C、HDL-C的含量;HE染色观察主动脉形态学变化;荧光定量PCR法检测主动脉中自噬相关因子Atg5、Beclin1和mTOR的mRNA表达水平。结果:与正常对照组相比,模型组TG、TC和LDL-C的表达升高,HDL-C表达降低;诱导了Atg5mRNA及Beclin1mRNA表达,抑制了mTOR mRNA的表达。与模型组相比,四妙勇安汤各剂量组和辛伐他汀组降低了TG、TC和LDL-C的表达,升高了HDL-C的表达;进一步诱导了Atg5mRNA及Beclin1mRNA表达,抑制了mTOR mRNA表达。结论:四妙勇安汤能够抑制家兔AS的形成,推测与其诱导自噬相关因子Atg5、Beclin1的表达,抑制mTOR的表达相关,这可能是四妙勇安汤抗AS的作用机制之一。 Objective:To investigate the effects of Si-Miao-Yong-An-Decoction on blood lipid biochemical parameters and autophagy-related factors Atg5,Beclin1,and mTOR mRNA expression in New Zealand atherosclerotic rabbits,and to clarify its anti-AS mechanism.Methods:Male New Zealand rabbits were randomly divided into normal control group,model group,Si-Miao-Yong-AnDecoction small,medium and large dose group and simvastatin group,6 in each group.Except the blank group,all other groups were established with AS models.After continuous administration,the model was administered continuously for 6 weeks.The content of TG,TC,LDL-C,HDL-C in serum was detected by an automatic biochemical analyzer,and the aorta was observed by HE staining.Morphological changes;Real-time PCR was used to detect the mRNA expression levels of autophagy-related factors Atg5,Beclin1 and mTOR in the aorta.Results:Compared with the normal control group,the expression of TG,TC and LDL-C in the model group increased,while the expression of HDL-C decreased;the expression of Atg5 mRNA and Beclin1 mRNA was induced,and the expression of mTOR mRNA was suppressed.Compared with the model group,each dose of Si-Miao-Yong-An-Decoction group and simvastatin group reduced the expression of TG,TC and LDL-C,and increased the expression of HDL-C;further induced Atg5 mRNA and Beclin1 mRNA expression repressed mTOR mRNA expression.Conclusion:Si-Miao-Yong-An-Decoction can inhibit the formation of AS in rabbits.It is speculated that the mechanism is related to the induction of autophagy-related factors Atg5 and Beclin1 and the inhibition of mTOR expression.This may be the mechanism of Si-Miao-Yong-An-Decoction against AS.
