甲状腺乳头状癌组织中USP33和CXCR4表达及其与JAK3、STAT3信号分子的关系 被引量:1

Expression of USP33 and CXCR4 in papillary thyroid carcinoma and its relationship with JAK3 and STAT3 signaling molecules

摘要 目的探讨PTC组织中USP33和CXCR4表达及其与JAK3、STAT3信号分子的关系。方法采用免疫组化染色EnVision法检测甲状腺乳头状癌(papillary thyroid carcinoma,PTC)组织中USP33及CXCR4的表达;采用Western blot技术检测6例PTC组织及癌旁组织中USP33的表达;采用qRT-PCR技术检测6例PTC组织及癌旁组织中USP33、CXCR4、JAK3及STAT3的表达。结果(1)免疫组化:USP33阳性表达位于细胞质,在肿瘤体积较大患者的PTC组织中表达量下降(P<0.05),CXCR4阳性表达位于细胞质,表达水平与远处转移、TNM分期有关(P<0.001);(2)Western blot:癌旁组织中USP33的表达水平高于PTC组织(P<0.05);(3)qRT-PCR:癌旁组织中USP33 mRNA在的表达略高于PTC组织(P>0.05),CXCR4 mRNA、JAK3 mRNA在PTC组织中的表达明显高于癌旁组织(P<0.05)。在PTC组织中,USP33及CXCR4蛋白表达的弱阳性率分别为42%,3.2%;中度阳性率分别为20%,38.7%;强阳性率为28%,32.3%。结论USP33、CXCR4和JAK3信号分子可能参与了PTC的发生、发展。 Objective To investigate the expression of USP33 and CXCR4 in PTC tissues and its relationship with JAK3 and STAT3 signaling molecules.Methods Immunohistochemical staining EnVision method was used to detect the expression of USP33 and CXCR4 in PTC tissues.Western blot was used to detect the expression of USP33 in 6 cases of PTC tissues and adjacent tissues.qRT-PCR was used to detect the expression of USP33,CXCR4,JAK3 and STAT3 in 6 cases of PTC tissues and adjacent tissues.Results(1)Immunohistochemistry:The positive expression of USP33 was located in the cytoplasm,and the expression level decreases in patients with large tumor volume(P<0.05).The positive expression of CXCR4 was located in the cytoplasm,The expression level was related to distant metastasis and TNM staging(P<0.001).(2)Western blot:The expression level of USP33 in tissues adjacent to cancer was higher than that in PTC tissues(P<0.05).(3)qRT-PCR:The expression of USP33 mRNA in tissues adjacent to cancer was slightly higher than that in PTC tissues(P>0.05),and the expression of CXCR4 mRNA and JAK3 mRNA in PTC tissues was significantly higher than that in adjacent tissues(P<0.05).In PTC tissues,the weak positive rates of USP33 and CXCR4 protein expression were 42%and 3.2%,the moderate positive rates were 20%and 38.7%,and the strong positive rates were 28%and 32.3%.Conclusion USP33,CXCR4 and JAK3 signaling molecules may be involved in the occurrence and development of PTC.
