摘要
目的通过高嘌呤饮食构建果蝇高尿酸血症(Hyperuricemia,HUA)模型,探讨尿酸(uric acid,UA)对果蝇爬行能力的影响。方法收集羽化后8h内的野生型黑腹果蝇(W1118)1360只,雌雄各半。将上述果蝇随机分为对照组、低浓度干预组、中浓度干预组和高浓度干预组四个实验组,每组340只果蝇,雌雄各170只。实验周期为7d,在实验结束后测定各实验组果蝇体内UA水平,以及果蝇生存率和爬行能力的变化。结果随着果蝇体内UA水平的不断升高,果蝇的生存率出现下降的趋势。尤其是高浓度干预组果蝇的生存率显著低于对照组,且高浓度干预组雄性果蝇的生存数量显著低于雌性果蝇,其差异均具有统计学意义(P<0.01);同时各干预组果蝇的爬行能力均出现不同程度的降低,中、高浓度干预组果蝇的爬行能力显著低于对照组,其差异具有统计学意义(P<0.05);相同干预条件下,高浓度干预组雄性果蝇的爬行能力显著低于雌性果蝇,其差异具有统计学意义(P<0.01)。结论高嘌呤饮食可以显著提高果蝇体内UA水平,并诱发果蝇运动障碍,提示UA可能通过诱发炎症反应影响果蝇的爬行能力。另外,雌激素在UA的体内代谢过程中扮演着重要角色。
Objective To investigate the effect of uric acid(UA)on the crawling ability of drosophila by using a model of Hyperuricemia(HUA)in drosophila established through hyperpurine diet.Methods Within 8 hours emergence,1360 wild-type fruit flies(W1118)were collected,half of male and female.The fruit flies were randomly divided into four experimental groups:a control group,a low-concentration intervention group,a medium-concentration intervention group,and a high-concentration intervention group,with 340 flies in each group(170 males and females).The experimental period was 7 days.After the experiment,the UA level of Drosophila melanogaster in each experimental group,and the changes of Drosophila survival rate and crawling ability were measured.Results With the continuous increase of UA levels in drosophila,the survival rate of drosophila tended to decline.In particular,the survival rate of fruit flies in the high-concentration intervention group was significantly lower than that of the control group,and the number of male drosophila in the high-concentration intervention group was significantly lower than that of female drosophila,and the difference was statistically significant(P<0.01).At the same time,the crawling ability of drosophila in each intervention group was reduced to varying degrees.The crawling ability of drosophila in the middle and high concentration intervention groups were significantly lower than the control group,and the difference was statistically significant(P<0.05).Under the same intervention conditions,the crawling ability of male drosophila in the high-concentration intervention group was significantly lower than that of female drosophila,and the difference was statistically significant(P<0.01).Conclusion High-purine diet can significantly increase UA levels in drosophila and induce dyskinesia in drosophila,suggesting that UA may affect the crawling ability of drosophila by inducing inflammatory response.In addition,estrogen plays an important role in the metabolism of UA in the body.
