摘要
目的观察自拟中药汤剂“湿疹溻渍方”外用对湿疹模型小鼠皮损中γ-干扰素受体(IFN-γRa)、白细胞介素-2(IL-2)、白细胞介素-4(IL-4)以及外周血中T淋巴细胞亚群CD4^+T、CD8^+T的影响。方法将7%的二硝基氯苯(DNCB)-丙酮溶液均匀涂抹于小鼠腹部脱毛的皮肤上,制作小鼠接触性湿疹模型,以中药汤剂湿疹溻渍方进行溻渍洗浴治疗,通过免疫组织化学法检测各个小组中小鼠腹部皮损组织中IFN-γRa、IL-2及IL-4的含量,采用流式细胞技术检测各组小鼠血浆中CD4^+T和CD8^+T的水平。结果“湿疹溻渍方”对DNCB致小鼠腹部湿疹有明显的改善效果,能够让湿疹小鼠皮损组织中的IFN-γRa和IL-2量增加,而IL-4的表达降低,且能够减小湿疹小鼠外周血中CD8^+T、升高CD4^+T淋巴细胞的百分比。结论“湿疹溻渍方”外用可以明显地改善小鼠湿疹模型,具体的作用原理与其参与免疫调节有关。
Objective To observe the effect of external application of self-formulated Shizhen Tazi Formula on the levels ofγ-interferon receptor(IFN-γRa),interleukin-2(IL-2)and interleukin-4(IL-4)and T lymphocyte subsets of CD4^+T and CD8^+T in the peripheral blood of the skin lesions of model mice with eczema.Methods A 7%dinitrochlorobenzene(DNCB)-acetone solution was applied to depilated abdominal skin of mice to establish the eczema model.Shizhen Tazi Formula was used to be drenched with sweat stains bath therapy,and immunohistochemistry(IHC)assay was used to measure the levels of IFN-γRa,IL-2,IL-4 in the abdominal lesion tissues of mice in all groups,and flow cytometry was used to measure the levels of CD4^+T and CD8^+T in plasma of mice in all groups.Results Shizhen Tazi Formula significantly improved DNCB induced abdominal eczema in mice,increased the levels of IFN-γRa and IL-2,reduced the expression level of IL-4,reduced the percentage of CD8^+T lymphocytes,and increased the percentage of CD4^+T lymphocytes in peripheral blood.Conclusion External application of Shizhen Tazi Formula can significantly improve the symptoms of model mice and the mechanism may be associated with its participation on immune regulation.
