摘要
目的探讨九香虫石油醚提取物(CcE)对锰中毒大鼠睾丸组织损伤的干预效果及Claudin1的作用。方法成年雄性SD大鼠50只,随机分为5个组:空白对照组、锰模型组、CcE低剂量干预组(50 mg/kg/d)、CcE中剂量干预组(100 mg/kg/d)、CcE高剂量干预组(200 mg/kg/d)。锰染毒建立大鼠生殖损伤模型,使用不同剂量的CcE进行干预,监测大鼠的行为学改变及生殖器官脏器系数和精子数目,并进行睾丸锰含量和形态学变化分析,检测Claudin1表达水平及分布,评价干预效果。结果锰模型组与空白对照组比较,大鼠的行为学指标即扑捉、骑跨、插入及射精的次数明显降低(P<0.05);CcE各剂量干预组与锰模型组比较,大鼠的上述行为学指标明显升高(P<0.05)。锰模型组与空白对照组比较,大鼠睾丸、附睾脏器系数与精子数目明显减少(P<0.05);CcE各剂量干预组与锰模型组比较,上述指标均有所升高。锰模型组与空白对照组比较,大鼠锰含量明显增高(P<0.05);CcE低剂量干预组与锰模型组比较,大鼠锰含量明显降低(P<0.05)。锰模型组与空白对照组比较,大鼠睾丸组织Claudin1蛋白的表达量明显降低(P<0.05);CcE中、高剂量干预组与锰模型组比较,Claudin1蛋白的表达量明显增加(P<0.05)。锰模型组与空白对照组比较,大鼠睾丸组织结构破坏,Claudin1分布减少且仅基底膜处可见;CcE各剂量干预组与锰模型组比较,大鼠睾丸组织结构明显改善,Claundin1分布增多且可见于各级精母细胞与支持细胞之间。结论九香虫石油醚提取物可拮抗锰引起的大鼠生殖系统损伤,且与Claudin1相关。
Objective To investigate the effect of petroleum ether extract of Coridius chinensis(CcE)on testicular tissue damage in rats with manganese poisoning and the role of claudin1.Methods Fifty adult male SD rats were randomly divided into five groups:blank control group,manganese model group,low dose intervention group(50 mg/kg/d),medium dose intervention group(100 mg/kg/d)and high dose intervention group(200 mg/kg/d).The reproductive damage model of rats was established by manganese exposure,and different doses of CcE were used for intervention.The behavioral changes,organ coefficient of reproductive organs and sperm number were monitored.The manganese content and histological changes of testis were analyzed.The expression level and distribution of claudin1 were detected.The activities of intervention were evaluated.Results The frequency of catching,riding,inserting and ejaculating in the manganese model group were significantly lower than those of the blank control group(P<0.05);the above indexes in each dose of CcE intervention group were significantly higher than those of the manganese model group(P<0.05).The organ coefficient of testis and epididymis and the number of sperm in the manganese model group were significantly lower than those of the blank control group(P<0.05);the above indexes in each dose of CcE intervention group were slightly higher than those of the manganese model group.The manganese content in the manganese model group was significantly higher than that of the blank control group(P<0.05);the content of manganese in the low dose intervention group was significantly lower than that of the manganese model group(P<0.05).The protein expression of claudin1 in testis of rats in manganese model group was significantly lower than that of the blank control group(P<0.05);the expression of claudin1 in the middle and high dose of CcE intervention group were significantly higher than that of the manganese model group(P<0.05).Compared with the blank control group,the testicular tissue structure of the manganese model group was destroyed,the distribution of claudin1 was decreased and observed in the basement membrane;compared with the manganese model group,the testicular tissue structure of the rats in each dose of CcE intervention groups were significantly improved,and the distribution of clandin 1 were increased and observed between spermatocytes and Sertoli cells.Conclusion The petroleum ether extract of Coridius chinensis can antagonize the damage of reproductive system induced by manganese which was related with claudin1.
