茶多酚对脂多糖诱导的人脐静脉内皮细胞中MCP-1、IL-6表达水平的影响 被引量:2

摘要 目的探讨茶多酚(Tea polyphenol,TP)对牙龈卟啉单胞菌脂多糖(Lipopolysaccharide,LPS)诱导的人脐静脉内皮细胞(Human umbilical vein endothelial cell,HUVEC)中单核细胞趋化蛋白-1(Monocyte chemoattractant protein-1,MCP-1)、白细胞介素-6(Interleukin-6,IL-6)表达水平的影响。方法分别选取一定浓度的TP和一定浓度的牙龈卟啉单胞菌LPS作用于人脐静脉内皮细胞,用ELISA法及Western blot法检测HUVEC中MCP-1、IL-6的表达水平。结果IL-6、MCP-1表达水平在阳性对照组(LPS)HUVEC中明显升高(P<0.05),其在TP预处理组HUVEC中的表达水平则明显降低(P<0.05);此外,IL-6、MCP-1蛋白表达水平也较LPS组显著下降(P<0.05)。结论TP可抑制牙龈卟啉单胞菌LPS诱导的HUVEC中MCP-1、IL-6表达水平上调,具有保护内皮细胞的作用。 Objective To explore the effect of tea polyphenol(TP)on the effect of monocyte chemoattractant protein-1(MCP-1)and interleukin-6(IL-6)expression levels in human umbilical vein endothelial cells(HUVEC)induced by 1 ipopolysaccharide(LPS).Methods Effects of various concentrations of TP on selected concentration of Porphyromonas gingivalis LPS in HUVEC were examined,and the expression levels of MCP-1 and IL-6 in HUVEC were detected by ELISA and Western blot.Results The expression levels of IL-6 and MCP-1 were significantly increased in LPS-treated HUVEC(P<0.05);TP pretreatment of HUVEC reduced the expression levels of IL-6 and MCP-1 by ELISA and protein compared with LPS group(P<0.05).Conclusion TP can inhibit the up-regulation of IL-6 and MCP-1 expression in HUVEC induced by Porphyromonas gingivalis LPS,and has the effect of protecting endothelial cells.
