摘要
君迁子是雌雄异株柿属植物,因其综合抗性较强,在生产上常用作柿子的砧木.君迁子早期性别鉴定对其实生后代的分类、管理与开发利用具有重要的作用.利用柿属植物中已发现的一个与雄性性别连锁的DISx-AF4S标记,对51株已知性别的君迁子性别进行验证,准确率可达98%以上,对113份未知性别的君迁子实生苗基因组DNA进行PCR扩增,有49份材料出现了目的条带,64份没有目的条带,说明供试的113份君迁子材料中有49株为雄株,其余为雌株.
Date plum(Diospyros Lotus L.),a dioecious persimmon plant,because of its strong comprehensive resistance,it is often used as the root stock of persimmon in production.The early sex identification of Date plum plays an important role in the classification,management,development and utilization in juvenile period.In this experiment,the male sex-linked DISx-AF4S marker,which was developed recently,was applied to verify the accuracy of 51 strain known sex gender Date plum,and the accuracy can reach more than 98%.Then the genomic DNA of 113 unknown sex gender seedlings was amplified by PCR,and the results showed that 49 materials appeared the target bands,64 materials showed no target bands,which indicated that 49 strains are male seedlings and the others are female.
