复合藻通过调节Stir3改善线粒体能量平衡抑制紫外线诱导的皮肤光老化 被引量:2

摘要 目的探索复合藻提取物对紫外线诱导的皮肤光老化的抑制作用及其分子机制.方法采用UVB诱导建立人上皮细胞衰老模型,给予不同浓度复合藻提取物处理人上皮细胞后建立模型,通过MTT实验考察细胞活力,DHE染色考察细胞氧化应激水平,检测细胞中ATP/ADP,NAD+/NADPH含量,线粒体复合物Ⅰ、Ⅱ、Ⅲ、Ⅳ活性,细胞色素C含量评价复合藻提取物对细胞线粒体功能及能量供应的影响.结果复合藻提取物干预后可以显著抑制UVB刺激对的人上皮细胞活力的影响,显著降低细胞ROS含量,中,高剂量组复合藻提取物干预可以显著升高ATP/ADP,NAD+/NADPH比值与模型组具有统计学差异(P<0.05),同时,高剂量复合藻可改善UVB对线粒体复合物Ⅰ、Ⅱ、Ⅲ、Ⅳ活性的抑制作用,且与模型组具有统计学差异(P<0.05),并减少细胞色素C含量.蛋白印迹实验结果表明,复合藻提取物可以显著升高细胞中Sirt3,Nrf2蛋白的表达.结论复合藻提取物可以通过上调Sirt3表达,激活Nrf2并提高线粒体复合酶活性,促进线粒体能量代谢水平,改善紫外线诱导的皮肤光老化. Objective To explore the inhibitory effect of compound algae on ultraviolet induced skin photoaging and its molecular mechanism.Methods The aging model of human epithelial cells was established by UVB induction,and the model was established after the treatment of human epithelial cells with different concentrations of compound algae.The cell viability was examined by MTT test,the oxidative stress level of cells was examined by DHE staining,the content of ATP/ADP,NAD+/NADPH,the activity of mitochondrial complexⅠ,Ⅱ,Ⅲ,Ⅳ,and cytochrome C content were used to evaluated the function of compound algae on cell mitochondria function and energy supply.Results The activity of human epithelial cells inhibited by UVB was significantly increased and the ROS content was significantly decreased after the intervention of compound algae.The ATP/ADP,NAD+/NADPH ratio were significantly higher than model group after pretreatment with middle and high dose compound algae(P<0.05).Meanwhile,the compound algae in high dose could improve the the activity of substance I,II,III and IV which inhibited by UVB and has statistically different from model group(P<0.05).The content of cytochrome C was also decreased.Western blotting results showed that compound algae could significantly increase the expression of SIRT3 and Nrf2 protein in cells.Conclusion compound algae can activate Nrf2 and improve the activity of mitochondrial complex enzyme,promote the level of mitochondrial energy metabolism,and improve UV induced skin photoaging by up regulating SIRT3 expression.
