白藜芦醇对肺癌A549细胞放疗的增敏效果及其与钙池操纵性钙内流的关系

Sensitization effect of resveratrol on radiotherapy of lung cancer A549 cell and its relationship with store-operated calcium entry

目的探究白藜芦醇(Res)对肺癌A549细胞放疗的增敏效果及其与钙池操纵性钙内流(SOCE)的关系。方法不同浓度Res联合不同剂量放射治疗(RT)处理A549细胞后,通过镜下观察和线粒体膜电位检测比较细胞增殖、凋亡情况,再通过钙成像技术和Western blot法检测SOCE及其关键蛋白的表达情况。结果与单一处理方式相比,RT+Res可以抑制A549细胞中的SOCE过程(P<0.05)。Res联合RT可以降低STIM1和Orai1蛋白在肿瘤细胞中的表达(P<0.01)。结论Res可以增强肺癌A549细胞的RT效果,而这种作用可能与抑制STIM1和Orai1介导的SOCE有关。

Objective To investigate the sensitization effect of resveratrol(Res)on radiotherapy of lung cancer A549 cell and its relationship with store-operated calcium entry(SOCE).Methods After A549 cells were treated with different concentration of Res combined with different doses of radiation therapy(RT),the cell proliferation and apoptosis were compared by microscopic observation and mitochondrial membrane potential detection.The SOCE and expression of the key proteins were detected by calcium imaging technique and Western blot method.Results Compared with the single treatment method,RT+Res can inhibited the SOCE process in A549 cells(P<0.05).Res combined with RT could reduce the expressions of STIM1 and Orai1 proteins in tumor cells(P<0.01).Conclusion Res can enhance the RT effect of lung cancer A549 cells,which may be related to the inhibition of STIM1 and Orai1-mediated SOCE.
