摘要 基于基因间隔序列(internal transcript space,ITS)分析中国12个不同地理居群的125份灰肉红菇(Russula griseocarnosa)样品的遗传多样性、居群遗传分化程度和遗传变异情况。结果表明:125份样品中检测到32个单倍型,不同地理居群的遗传距离与地理距离之间没有对应关系,二者无明显相关性;同一地理居群内单倍型多样性较高,核苷酸多样性较低;整体的遗传分化指数F_(st)为0.59955,基因流N_m为0.17,这些结果表明125份灰肉红菇样品基因交流水平低,分化程度较高;分子方差分析(analysis of molecular variance,AMOVA)结果表明,居群间遗传变异占总变异的22%,居群内的遗传变异占总变异的78%,因此遗传分化主要来自于居群内部。 To investigate the genetic diversity and differentiation of Russula griseocarnosa population in China,we analyzed the molecular variation,genetic diversity and population genetic differentiation of 125 samples from 12 different geographical populations in China based on their ITS sequences.The results showed that 32 haplotypes were detected in the 125 ITS sequences.There was no significant correlation between the genetic distance of each geographical population and the geographical distance of each geographical population.The haplotype diversity of each geographical population was high and the nucleotide diversity was low.The overall genetic differentiation index(Fst)was 0.59955 and the gene flow(Nm)was 0.17,suggesting that the 125 samples were highly diverse with a low level of gene exchange.The analysis of molecular variance(AMOVA)showed that the genetic variation among and within populations accounted for 22%and 78%of the overall variance respectively,suggesting that genetic differentiation mainly happened within populations.
