基于形态特征和多基因联合序列鉴定卵孢长根菇蛛网病病原菌 被引量:5

Identification of a Pathogenic Fungus Causing Cobweb Disease in Oudemansiella raphanipes by Morphological Characteristics and Phylogenetic Analysis

摘要 从感染病害的卵孢长根菇(Oudemansiella raphanipes)子实体中获得1株真菌CG01,采用平板培养观察该菌的菌落及镜检其菌丝、分生孢子梗、分生孢子等形态特征,采用ITS、RPB1、RPB2及TEF序列对其进行系统发育分析,另外通过柯赫氏法则验证其致病性。此外研究不同温度(15、20、25、30、35℃)对该菌生长的影响。结果表明:引起卵孢长根菇蛛网病的病原菌是与Cladobotryum paravirescens及C.asterophorum亲缘关系较近的葡枝霉属(Cladobotryum)真菌,病原菌菌丝体的最适生长温度为25℃,本研究首次鉴定出引起卵孢长根菇蛛网病的病原菌。 In order to identify pathogenic fungi causing cobweb disease in Oudemansiella raphanipes,a diseased fruiting body was subjected to tissue isolation and single spore isolation to obtain a pathogenic strain CG01.CG01 was then identified by its morphological characteristics and multigene phylogenetic analysis.For the morphological identification,CG01 was observed for its colony characteristics on PDA medium,and its microscopic characteristics of hyphae,conidiophores and conidia.For the phylogenetic analysis,sequences of ITS,RPB1,RPB2 and TEF were used.Based on the results of the above analyses and the Koch’s postulates,CG01 was confirmed to be a strain of Cladobotryum sp.,with rather close consanguinity with C.paravirescens and C.asterophorum at the molecular level.The optimal temperature for the hyphal growth of CG01 was found to be 25℃.This is the first report of pathogen responsible for cobweb disease in O.raphanipes.
