摘要
目的研究上皮细胞转化因子2(ECT2)在胰腺癌转移的作用及其可能的诱导表皮生长因子受体(EGFR)相关的上皮-间充质转化(EMT)机制。方法使用免疫组织化学和Western blotting方法检测ECT2在胰腺癌组织及细胞系中的表达情况。在胰腺癌细胞中降低ECT2的表达,通过细胞划痕实验测定胰腺癌的细胞迁移变化,使用Transwell实验检测胰腺癌细胞的侵袭力。通过Western blotting方法测定EMT标志物和EGFR的表达变化。结果ECT2在胰腺癌组织和细胞中显著上调,并提示不良预后。ECT2调控胰腺癌细胞的侵袭和迁移。ECT2沉默下调EGFR表达同时抑制EMT。结论ECT2可能通过EGFR信号通路相关的EMT促进胰腺癌细胞的侵袭性。
Objective To investigate the role of epithelial cell transforming factor 2(ECT2)in pancreatic cancer metastasis and its possible mechanism of inducing epidermal growth factor receptor(EGFR)mediated epithelial-mesenchymal transition(EMT).Methods Expression of ECT2 in pancreatic cancer tissue and pancreatic cancer cell lines were characterized using immunohistochemistry and Western blotting.ECT2 was knocked down in pancreatic cancer cells,then the cell migration of pancreatic cancer was measured by wound healing test,and the invasion of pancreatic cancer cells was detected using the Transwells.Expression of markers of EMT and EGFR were assayed by Western blotting.Results ECT2 was significantly upregulated in pancreatic cancer tissues and cells,suggesting a poor prognosis.ECT2 regulated the invasion and migration of pancreatic cancer cells in vitro.ECT2 silencing downregulated EGFR expression and inhibited EMT.Conclusion ECT2 may promote the invasion of pancreatic cancer cells through EGFR signaling pathway related EMT.
