辣椒素激活TRPV1改善大鼠功能性排便障碍的机制研究

摘要 为探讨瞬时感受器电位香草素受体1(TRPV1)激动剂辣椒素对功能性排便障碍(FDD)大鼠排便的影响及机制,本研究将40只SD大鼠随机分为4组,即正常对照组、辣椒素组、FDD模型组、FDD+辣椒素组,进行实验研究。观察各组大鼠排便情况,采用免疫组化法检测TRPV1在大鼠直肠组织中的表达情况,通过Western Blot检测大鼠直肠组织中TRPV1、P物质、血管活性肠肽(VIP)的蛋白表达水平,并进行对比分析。结果显示,FDD+辣椒素组大鼠排便情况较FDD组好转。免疫组化结果显示,FDD组、FDD+辣椒素组、正常对照组、辣椒素组大鼠直肠组织中TRPV1免疫阳性着色细胞逐步增多且染色逐渐加深。Western Blot结果显示,与正常对照组及辣椒素组相比,FDD组大鼠直肠组织中TRPV1及P物质水平下降、VIP水平升高(均P<0.05);与FDD组相比,FDD+辣椒素组TRPV1水平升高、VIP水平下降(均P<0.05)。相关性分析显示,TRPV1与P物质水平无相关性(r=0.046,P>0.05),TRPV1、P物质水平与VIP水平均呈负相关(r=-0.604、r=-0.329,P<0.05)。结果表明,辣椒素对FDD大鼠的排便具有改善作用,其机制可能与辣椒素调节TRPV1、P物质、VIP蛋白的表达水平有关。 This research was to study the role and mechanism of capsaicin activating transient receptor potential vanilloid 1(TRPV1)for improving rats’functional defecation disorder(FDD),randomly divided 40 SD rats into four groups:normal controlled group(NC group),capsaicin group(C group),FDD model group(FDD group),and(FDD+capsaicin)group(FC group),as well performed on them following items as observing their defecation status,detecting TRPV1 expression in their rectal tissue by using immunochemical method(IHC);and,detecting and comparing the expression levels of TRPV1,P substance,and vasoactive intestine peptide(VIP)by using Western Blot method.As results,the defecation status of FC group got improved than that of FDD group;IHC detection resutls:in order of FDD group,FC group,NC group and C group rats’TRPV1 immuno-staining positive cells became more,and their colour deeper;Western Blot detection results:compared with NC group and C group,FDD group’s TRPV1 and P substance levels in rectal tissue decreased,but VIP level increased,meanwhile,compared with FDD group,FC group’s TRPV1 level increased,but VIP level decreased(all,P<0.05);As well,relativity analysis results:TRPV1 level was not related to P substance level(r=0.045,P>0.05),but TRPV1 and P substance level were negatively related to VIP level(r=-0.604,r=-0.329,P<0.05).Results show that capsaicin has the role for improving FDD rats’defecation and the mechanism could be related to its regulating the expression levels of TRPV1,P substance and VIP protein.
