摘要
目的通过佐剂性关节炎大鼠模型,筛选出长柱金丝桃抗类风湿性关节炎的活性部位。方法制备长柱金丝桃石油醚、乙酸乙酯、正丁醇、水部位提取物,参照经典急性毒性实验方法确定长柱金丝桃不同提取部位的急性毒性;将佐剂性关节炎模型大鼠,随机分成7组:正常对照组、模型对照组、阳性对照组(雷公藤多苷)、石油醚部位组、乙酸乙酯部位组、正丁醇部位组、水部位部位组,每组8只。分组给药24 d,观察测定足肿胀度、关节炎评分指数、免疫器官(胸腺、脾脏)指数的变化。结果长柱金丝桃乙酸乙酯部位、正丁醇部位半数致死量(LD50)分别为22.13 g·kg^-1、31.37 g·kg^-1;水部位、石油醚部位最大给药量(MLD)分别为58.66 g·kg^-1、60.12 g·kg^-1;与模型组相比,乙酸乙酯部位给药组大鼠足肿胀度及关节肿胀指数显著降低(P<0.01);胸腺指数、脾脏指数均显著升高(P<0.01)。结论长柱金丝桃对大鼠佐剂性关节炎治疗作用活性部位为乙酸乙酯提取部位。
Objective To establish an adjuvant arthritis rat model and screen the anti rheumatoid arthritis active site of Hypericum ascyron L.Methods The extracts of petroleum ether,ethyl acetate,n-butanol and water were prepared.The acute toxicity of different parts of Hypericum ascyron L.was studied by referring to the classical acute toxicity experiment method.The adjuvant arthritis model rats were randomly divided into 7 groups:normal control group,model control group,positive control group(Tripterygium wilfordii glycoside),petroleum ether part group and ethyl acetate part.Group B,n-butanol group and water group,8 rats in each group.The changes of swelling degree of foot,index of arthritis score and index of immune organs(thymus and spleen)were observed.Results The LD50 of ethyl acetate and n-butanol were 22.13 g·kg^-1 and 31.37 g·kg^-1,respectively,and the MLD of water and petroleum ether were 58.66 and 60.12 g·kg^-1.Compared with the model group,the swelling degree of foot and joint in the ethyl acetate group were significantly reduced(P<0.01),the thymus index and spleen index were significantly increased(P<0.01).Conclusion The active part of Hypericum ascyron L.in the treatment of adjuvant arthritis in rats is ethyl acetate extract.
