辣椒炭疽zf-C2H2基因家族的生物信息学分析 被引量:2

摘要 C2H2型锌指(zinc finger, zf)蛋白具有识别与结合特异性DNA片段等功能,参与基因表达的调控。编码zf-C2H2蛋白的基因以基因家族的形式存在。本研究通过比对已开发的辣椒炭疽基因组数据查找到19个zf-C2H2基因家族成员,对其理化性质、亚细胞定位、系统进化、蛋白质的二级结构、蛋白质的三级结构、保守基序进行分析。通过理化性质分析,发现辣椒炭疽zf-C2H2蛋白多为碱性蛋白,氨基酸数目在386-985aa,蛋白质等电点在6.05-10.04,分子量在42002.8-109661.9KD,亚细胞定位表明zf-C2H2蛋白全部定位于细胞核中。根据系统进化关系可分为7个亚族,第4亚族和第7亚族基因的数量为2,其余的为3。通过对辣椒炭疽zf-C2H2蛋白的二级和三级结构分析,发现zf-C2H2蛋白主要以无规卷曲为主,各个成员的二级结构组成的排列顺序为无规卷曲>α-螺旋>延伸链>β-折叠。根据保守基序分析,发现辣椒炭疽zf-C2H2蛋白的序列共含有8个保守基序,其中motif1在19条序列中都存在。研究结果为后续辣椒炭疽zf-C2H2蛋白的深入分析提供一定的科学依据。 The C2H2 type zinc finger(zf)protein has the functions of recognizing and binding specific DNA fragments,and is involved in the regulation of gene expression.The genes encoding zf-C2H2 protein exist in the form of gene families.This study found 19 members of the zf-C2H2 gene family by comparing the genomic data of the developed Colletotrichum gloeosporioides.Their physical and chemical properties,subcellular localization,phylogenetic evolution,protein secondary structure,protein tertiary structure,and conserved motifs For analysis.Through physical and chemical properties analysis,it was found that the zf-C2H2 protein of Colletotrichum gloeosporioides was mostly basic protein,the number of amino acids was between 386-985aa,protein isoelectric point was between 6.05-10.04,molecular weight was between 42002.8KD-109661.9KD,subcellular Localization showed that the zf-C2H2 protein was all located in the nucleus.According to the phylogenetic relationship,it can be divided into 7 subfamilies.The number of genes in the 4th and 7th subfamilies is 2,and the rest are 3.By analyzing the secondary and tertiary structure of the zf-C2H2 protein of Colletotrichum gloeosporioides,it was found that the zf-C2H2 protein is mainly dominated by random coil,and the order of the secondary structure composition of each member is random coil>α-helix>extension Chain>β-sheet.According to the analysis of conserved motifs,it was found that the sequence of Colletotrichum gloeosporioides zf-C2H2 protein contains a total of 8 conserved motifs,among which motif1 is present in 19 sequences.The research results provide a certain scientific basis for the in-depth analysis of zf-C2H2 protein in Colletotrichum gloeosporioides.
