摘要
目的:研究17β-雌二醇(E2)对人脐静脉内皮细胞(HUVECs)增殖、迁移、侵袭和成管的影响。方法:不同浓度(10^-10、10^-9、10^-8、10^-7、10^-6 mol/L)的E2与HUVECs共培养,并设置空白对照组,用CCK-8法检测HUVECs的细胞活力,划痕实验检测HUVECs的迁移功能,Transweel实验检测HUVECs的侵袭功能,成管实验检测HUVECs的成管功能。结果:与空白对照组相比,浓度为10^-8 mol/L的E2组明显促进HUVECs的增殖、迁移、侵袭和成管,浓度为10^-6 mol/L的E2组明显抑制了HUVECs的增殖、迁移、侵袭和成管。结论:E2对于HUVECs的保护作用呈剂量效应关系,随着E2浓度升高,其促进HUVECs的增殖、迁移、侵袭和成管作用增强,但高浓度时则具有抑制效应。
Objective:To investigate the effects of 17β-estradiol(E2)on the proliferation,migration,invasion and tube formation of human umbilical vein endothelial cells(HUVECs).Methods:HUVECs were co-cultured by E2 in different concentrations(10^-10,10^-9,10^-8,10^-7,or 10^-6 mol/L),and blank control group was set up.The CCK-8 was used to detect the viability of HUVECs,the scratch test was used to detect the migration function of HUVEC,the transweel test was used to observe the invasion function of HUVEC,and the tube formation test was used to evaluate the tube formation function of HUVEC.Results:Compared to those of blank control group,10^-8 mol/L of E2 promoted HUVECs proliferation,migration,invasion,and tube formation significantly,but 10^-6 mol/L of E2 inhibited HUVEC proliferation,migration,invasion,and tube formation significantly.Conclusion:The protective effect of E2 on HUVECs has the relationship of dose-effect.As the concentration of E2 increases,E2 can promote the proliferation,migration,invasion and tube formation of HUVECs,while when the concentration of E2 is high,it has an inhibitory effect.
