新型超微量冷冻载体在人少精子冻融中的应用价值 被引量:2

摘要 目的:探讨新型超微量冷冻载体在人稀少精子冻融中的应用效果。方法:收集2018年6—12月本院因男方严重少弱精子症在女方取卵日行卵胞质内单精子注射(ICSI)治疗的患者30例,分别用新型超微量冷冻载体和超微量冷冻麦管对患者10条活动精子进行快速冷冻,比较两种载体不同冷冻体积的精子回收率、活动率和存活率,并用新型超微量冷冻载体冻融存活的精子与体外培养成熟的卵子行ICSI,比较新鲜精子与冷冻精子的受精率和卵裂率。结果:新型超微量冷冻载体0.5μl、1μl和2μl组冷冻液量解冻后精子回收率(94.3%、95.0%、94.0%)、活动率(45.2%、44.9%、42.9%)及存活率(65.7%、64.2%、62.7%)比较无差异,超微量冷冻麦管5μl和10μl组冷冻液量解冻后精子回收率(61.7%、57.0%)、活动率(39.5%、37.4%)及存活率(53.5%、52.6%)比较无差异(均P>0.05);但新型超微量冷冻载体2μl组的精子回收率(94.0%)、存活率(62.7%)高于超微量冷冻麦管5μl组(61.7%、53.5%)(P<0.05)。新型超微量冷冻载体复苏组存活精子ICSI受精率(77.8%)、卵裂率(96.8%)与新鲜精子组(82.8%、99.1%)比较无差异(P>0.05)。结论:新型超微量冷冻载体可作为人稀少精子冻融的有效载体,有良好的应用价值。 Objective:To evaluate the application effect of new micro-cryopreservation carrier in sperm cryopreservation of man with oligo-asthenospermia spermatozoa.Methods:10 active spermatozoa from each semen sample of 30 men with severe oligoasthenospermia were frozen by micro-straw and new micro-cryopreservation carrier when their wives had accepted intracytoplasmic sperm injection(ICSI)on the day of the retrieval from June 2018 to December 2018.The sperm recovery rate,sperm motility rate,and sperm viability rate were compared between micro-straw and new micro-cryopreservation carrier with different freezing volumes.After the mature eggs cultured in vitro were conducted ICSI,the fertilization rate and cleavage rate of their wives were compared between the fresh sperm and the frozen sperm by new micro-cryopreservation carrier.Results:The rate of sperm recovery of men after micro-cryopreservation with new freezing carrier volume in 0.5μl,1μl and 2μl were 94.3%,95.0%,and 94.0%,respectively,which had no significant different among them.The sperm motility of men after micro-cryopreservation with new freezing carrier volume in 0.5μl,1μl and 2μl were 45.2%,44.9%,and 42.9%,respectively,which had no significant different among them.And the sperm survival rate of men after micro-cryopreservation with new freezing carrier volume in 0.5μl,1μl and 2μl were 65.7%,64.2%,and 62.7%,respectively,which had no significant different among them.The rate of sperm recovery of men after micro-cryopreservation with freezing carrier volume in 5μl and 10μl were 61.7%and 57.0%,which had no significant different between them.The sperm motilities after micro-cryopreservation with freezing carrier volume in 5μl and 10μl were 39.5%and 37.4%,which had no significant difference between them.And the sperm survival rate of men after micro-cryopreservation with freezing carrier volume in 5μl and 10μl were 53.5%and 52.6%,which had no significant different between them(all P>0.05).The rates of sperm recovery and survival after micro-cryopreservation with new freezing carrier volume in 2μl were 94.0%and 62.7%,which were significant higher than those(61.7%and 53.5%)after micro-cryopreservation with freezing carrier volume in 5μl(P<0.05).The rates of fertilization and cleavage of survival sperm for ICSI after micro-cryopreservation with new freezing carrier were 77.8%and 96.8%,which had no significant different from those(82.8%and 99.1%)of fresh sperm(P>0.05).Conclusion:The new micro-cryopreservation carrier is better for oligo-asthenospermia spermatozoa cryopreservation,which has good application value.
