摘要 目的:研究雷公藤内酯醇(TP)用于荷卵巢癌大鼠的抑瘤效应及其对免疫系统的影响。方法:选取性SD大鼠作为实验动物,皮下注射卵巢癌细胞后得到荷卵巢癌大鼠,分为腹腔注射生理盐水的对照组、低剂量TP组、中剂量TP组、高剂量TP组。药物干预后测定肿瘤的质量和体积、肿瘤组织中抑癌基因的表达量、外周血中免疫细胞因子的水平。结果:低剂量、中剂量、高剂量TP组大鼠的肿瘤质量与体积、外周血中白细胞介素-4(IL-4)、白细胞介素-17(IL-17)、转化生长因子β1(TGF-β1)的水平均明显低于对照组,肿瘤组织内PTEN、ST7L、Bax、TIMP1、E-cadherin的mRNA表达量以及外周血中干扰素-γ(IFN-γ)、肿瘤坏死因子-α(TNF-α)的水平均明显高于对照组且TP剂量越大,肿瘤质量与体积、外周血中IL-4、IL-17、TGF-β1的水平越低,肿瘤组织内PTEN、ST7L、Bax、TIMP1、E-cadherin的mRNA表达量以及外周血中IFN-γ、TNF-α的水平越高。结论:TP用于荷卵巢癌大鼠具有显著的抑瘤效应,能够抑制肿瘤生长、增加抑癌基因表达并调节抗肿瘤免疫应答。 Objective:To study the antitumor effects of Triptolide(TP)used in rats with ovarian cancer and its influence on the immune system.Methods:Female SD rats were selected as experimental animals,and ovarian cancer-bearing rats were obtained after subcutaneous injection of ovarian cancer cells.They were divided into a model group,a low-dose TP group,a middle-dose TP group,and a high-dose TP group injected with physiological saline.After drug intervention,the weight and volume of the tumor,the expression of tumor suppressor genes in the tumor tissue,and the content of immune cytokines in the peripheral blood were measured.Results:Tumor weight and volume,interleukin-4(IL-4)in peripheral blood,interleukin-17(IL-17),and transforming growth factorβ1(TGF-β1)in rats in the low-dose,middle-dose,and high-dose TP groups were significantly lower than the model group,and the expression of PTEN,ST7L,Bax,TIMP1,E-cadherin mRNA in tumor tissues and interferon-γ(IFN-γ)and tumor necrosis factorα(TNF-α)in peripheral blood were significantly higher than the model group.The greater the TP dose,the lower the tumor weight and volume,and the content of IL-4,IL-17,and TGF-β1 in the peripheral blood,the higher the mRNA expression of PTEN,ST7L,Bax,TIMP1,E-cadherin in the tumor tissue,and the higher the content of IFN-γand TNF-αin peripheral blood.Conclusion:TP has a significant tumor suppressing effects on ovarian cancer-bearing rats,which can inhibit tumor growth,increase tumor suppressor gene expression and regulate anti-tumor immune response.
