摘要
目的:观察血清淀粉样蛋白A1(SAA1)和转化生长因子-β1(TGF-β1)在敌草快(DQ)中毒小鼠急性肺损伤中的表达及意义。方法:将128只SPF级健康雌性C57小鼠随机分为对照组(N组,n=32)和模型组(DQ组,n=96),模型组予实验首日一次性腹腔分别注射DQ 20、30、40 mg/kg复制中毒小鼠模型,对照组予注射等剂量生理盐水,常规饲养。于模型建立后第1、3、5、7天分4批次处死小鼠并取材。检测小鼠肺湿/干重比,苏木精-伊红染色观察肺组织病理变化,酶联免疫吸附法检测小鼠血清SAA1的表达情况,免疫荧光定量PCR法检测小鼠肺组织TGF-β1 mRNA表达的情况。结果:与对照组相比,模型组小鼠肺组织炎症损伤严重,血清SAA1含量明显增多(P<0.05);肺组织TGF-β1 mRNA的表达均增高(P<0.05)。结论:SAA1和TGF-β1在DQ中毒小鼠肺损伤中显著增高,参与其炎症过程,并可能在其病程进展中发挥重要作用。
Objective:To observe the expression and significance of serum amyloid A1(SAA1)and transforming growth factor-β(TGF-β1)in acute lung injury induced by diquat in mice.Method:One hundred and twenty-eight healthy SPF female C57 mice were randomly divided into a control group(N group,n=32)and a poisoning group(DQ group,n=96).The DQ group was given a single intraperitoneal injection of different dose of diquat on the first day of the experiment(20 mg/kg,30 mg/kg,40 mg/kg)to replicate the infected mouse model,and the control group was injected with an equal dose of normal saline for routine feeding.The mice were sacrificed in four batches on day 1,3,5,and 7 after the model was established and materials were collected.Detection of lung wet/dry weight ratio of mice,hematoxylin-eosin staining to observe the pathological changes of lung tissue,enzyme-linked immunosorbent assay to detect the expression of SAA1 in mice serum,immunofluorescence quantitative PCR method to detect TGF-β1 in mouse lung tissue mRNA expression.Result:Compared to the control group,the inflammatory injury of the lung tissue in the poisoned group was severer,and the serum SAA1 content was significantly increased(P<0.05).The expression of TGF-β1 mRNA in lung tissue was increased(P<0.05).Conclusion:SAA1 and TGF-β1 are significantly increased in lung injury induced by DQ mice,participately in the inflammatory process,and play an important role in the progression of the disease.
