青稞根际高效纤维素分解菌的分离及筛选 被引量:2

摘要 为获得青稞根际高效纤维素降解菌,充分利用西藏地区青稞等植物废渣中的纤维素,本实验经过CMC-Na培养基纯培养,刚果红染色,NaCl脱色的初筛法结合纤维素分解菌CMC酶活力测定的复筛法,从青稞根际土壤中筛选出酶活性较高的菌株,并对它们进行初步研究。通过初筛和复筛,从青稞根际土壤中共筛选出酶活性较强的真菌5株:P-3-1、P-3-2、P-5-2、P-6-1、P-8-1和2株细菌L-12-1和L-14-2,其中细菌菌株L-14-2酶活性最强,为368.06 U/mL。结果表明青稞根际土壤中具有分解能力较强的纤维素分解菌。获得青稞根际高效纤维素分解菌,对充分利用青稞等植物废渣具有重要意义,在西藏地区农业生产上具有良好的应用前景。 In order to obtain highly efficient cellulose degrading bacteria in barley rhizosphere,the cellulose in the waste residue of highland barley and other plants in Tibet should be fully utilized.In this experiment,through the pure culture of CMC Na medium,the dyeing of Congo red,the primary screening method of NaCl decolorization and the secondary screening method of CMC enzyme activity determination of cellulose decomposing bacteria,the strains with higher enzyme activity were screened out from the soil containing bacteria,which were preliminarily studied.Five fungal strains p-3-1,p-3-2,p-5-2,p-6-1,p-8-1 and two bacterial strains l-12-1 and l-14-2 with strong enzyme activity were screened out from the soil containing bacteria.Among them,the bacterial strain l-14-2 has the strongest enzyme activity(368.06 U/ml).The results showed that there were cellulose decomposing bacteria with strong decomposing ability in barley rhizosphere soil.It is of great significance to obtain highly efficient cellulose decomposing bacteria in barley rhizosphere for making full use of barley and other plant residues,and it has a good prospect in agricultural production in Tibet.
