摘要
目的:通过浸泡法使氯化镁(MgCl2)与组织工程牛心包片充分结合并进行动物实验,探索该种补片的各种性能,以期制备出具有良好机械性能、生物相容性及抗钙化的组织工程补片,从而更好的服务于临床。方法:采用Triton X-100及DNA/RNA核酶法脱细胞、明胶再基质化、EDC/NHS交联处理牛心包,采用浸泡法使MgCl2与经过上述处理的心包结合并进行大鼠皮下包埋,对各组补片进行机械性能、HE染色及钙镁含量测定等相关测试。结果:MgCl两组在机械强度上与去细胞组和交联组无明显差异,组织浸润较轻且短期内钙含量较其他两组明显减少。结论:使用MgCl2浸泡法处理的牛心包,在维持机械性能及组织学性能的基础上具有一定的抗钙化作用,为以后进一步的研究奠定了理论和实验基础。
Objective: Tissue engineered bovine pericardium tablets were first fully combined with magnesium chloride(MgCl2) and then tested in animals. By exploring the various properties of this patch, we hope to make a tissue engineering patch with good mechanical properties, biocompatibility, low immunogenicity and anti-calcification, so as to better apply it in clinic. Methods: From January 2005 to December 2005, the clinical data of 230 patients who underwent OPCABG and successfully discharged from the same operation group in Anzhen Hospital were analyzed and followed up. Results: The bovine pericardium was treated with Triton x-100, DNA/RNA ribozyme, gelatin, EDC/NHS, then MgCl2 was combined with the above pericardium, and finally subcutaneous implantation was performed in rats. The mechanical properties, HE staining, immunogenicity and calcium and magnesium content of each patch were tested. SPSS 20.0 was used for statistical analysis of the data. P<0.01 means the difference is statistically significant. Results: There was no significant difference in mechanical strength between the MgCl2 group and the cell-free group and the crosslinking group. The tissue infiltration in the MgCl2 group was lighter and the calcium content in the short term was significantly reduced compared with the other two groups. Conclusions: The bovine pericardium treated with MgCl2 immersion method has a certain anti-calcification effect on the basis of maintaining mechanical and histological properties, which lays a theoretical and experimental foundation for further research.
