摘要
以香豆素为母体,丙烯酸酯为识别基团,设计、合成了荧光探针3-(2-苯并噻唑基)香豆素-7-丙烯酸酯(探针1),用于高灵敏、选择性检测活细胞中的还原型谷胱甘肽(GSH)。探针1与GSH反应后,溶液由无色变为亮绿色,荧光强度与GSH浓度在0~10μmol/L范围内呈现良好的线性关系,检出限为74 nmol/L;此外,探针还具有选择性高、抗干扰能力强和稳定性好等优点。荧光成像实验表明,探针1具有良好的细胞膜通透性、低毒性和生物相容性,可用于细胞内GSH浓度的快速检测。
Coumarin was utilized as matrix with a receptor unit of acrylate to design and synthesize a fluorescent probe 3-(benzo[d]thiazol-2-yl)-2-oxo-2 H-chromen-7-yl acrylate(probe 1)for the highly sensitive and selective determination of reduced glutathione(GSH)in living cell.Once the GSH reacts with the probe 1,a colorimetric response from colorless to light green was produced.The fluorescence intensity and GSH concentration showed a good linearity in the range of 0-10 mol/L,with the detection limit of 74 nmol/L,indicating that probe 1 was highly sensitive,excellent selectivity and good stability to GSH.Fluorescence bioimaging assays proved that probe 1 owns excellent cell membrane permeability,low cytotoxicity and satisfied biocompatibility,which was desirable for fast detection and imaging of both endogenous and exogenous GSH in the living cells.
