整合型设计性综合大实验在基因工程教学中的应用与探索 被引量:1

Development of a novel comprehensive and integrated Gene Engineering laboratory practice program for scientific innovation cultivation

摘要 基因工程是生物制药专业的核心专业课,具有较强的理论性和实践性。基因工程实验教学是基因工程教学的核心环节,但目前普遍存在实验内容陈旧、教学方法枯燥、单元衔接散乱、演示性实验偏多等缺点,极大地降低了基因工程实验教学的效果。为了提高学生的实验技能和创新能力,我们提出了整合型设计性基因工程综合大实验的教学模式。以重组人β-catenin原核表达和分离纯化为核心内容,从实验准备、实验设计、实验操作和实验分析等方面,对基因工程实验教学模式进行了改革和探索。实践证明,整合型设计性基因工程综合大实验建立了从"分子设计到产品研发"的自由实践教学模式,显著提高了基因工程的教学质量,提高了学生的实验技能和综合素质,是培养学生科研思维和创新能力的有效途径。本文探索的整合型设计性基因工程综合大实验具有良好的推广性和示范性,可为基因工程教学提供有益的参考。 Gene Engineering is a key and compulsory course for biopharmaceuticals, which strongly integrates theory and practice. As a core content of teaching process, the laboratory practice program has been currently labeled with some prevalent defects, including the old-fashioned contents, dull teaching methods, scattered chapter joints and excess of demonstrations, which would greatly weaken the quality of laboratory practice training. For the urgent promotion needs in practice and scientific innovation cultivation, we have developed a comprehensive and integrated Gene Engineering laboratory practice program based on the bacterial expression and purification of recombinant human β-catenin. Then a novel laboratory practice program has been performed, which is featured by basic preparing process of biologicals from molecular cloning to targeted protein purification. As a result, this ongoing laboratory practice program has greatly improved the students’ basic skills, scientific passion and innovation. This novel comprehensive and integrated Gene Engineering laboratory practice program deserves to be popularized for scientific innovation cultivation, and could provide useful suggestions for future Gene Engineering teaching.
