纤维素链霉菌AU-10源脂肪酶 --一种可应用于洗涤剂配方的酶

摘要 酶的加入可以显著降低洗涤剂中合成化学物添加量和洗涤温度,从而降低洗涤剂产品的环境负担和经济负荷.本研究利用两相分离法从纤维素链霉菌AU-10菌株中提取获得了一种碱性脂肪酶,酶活回收率为68%,纯化倍数为7.08.这种纤维素链霉菌AU-10源脂肪酶在pH=9.040℃下表现出最高的酶活力.在pH 9.0~11.0之间时100%的酶活力可以维持1 h左右.该酶在30~50℃范围均可以保持较高的稳定性.它的最大反应速率是0.83 mmol·L-1·min-1,米氏指数为0.34 mmol·L-1.对橄榄油和葵花籽油有非常高的催化水解活性.从EDTA对酶活力的影响可以判定纤维素链霉菌AU-10源脂肪酶是一种金属酶,亚铁离子、铜离子和含硼化合物可以显著提高它的催化活性.无论是表面活性剂还是氧化剂,都不会对其稳定性产生明显影响.在市售的奥妙、碧浪、Tursil,Pril和Fairy中,该脂肪酶的活性依然可以达到108.8%、115.6%、98.35%、140.4%和107.6%.简言之,纤维素链霉菌AU-10源脂肪酶是一种可应用于洗涤剂配方的酶. Enzymes can decrease the environmental and economic load of detergent products by reducing the amount of chemicals used in detergents and by allowing washing at ambient temperatures.In this study,Streptomyces cellulosae AU-10(Gen Bank accession number:MG780240)lipase was purified 7.08-fold with 68%yield using an aqueous 2-phase system.The Streptomyces sp.AU-10 lipase showed maximal activity at pH 9.0 and 40℃.Hundred percent activities were measured in the pH range from 9.0 to 11.0 for 1h.The enzyme was also highly stable at 30~50℃.The values of K m and V max were calculated as 0.34 mM and 0.83 mM·min-1,respectively.The lipase has high hydrolytic activity for olive oil and sunflower oil.The effect of ethylenediamine tetraacetic acid on the enzyme has shown that the lipase is a metalloenzyme.The activity increased in the presence of Fe2+,Cu2+,and various boron compounds.The enzyme has shown a good stability not only with surfactants but also with oxidizing agents.In addition,activities in the presence of Omo,Ariel,Tursil,Pril,and Fairy were measured as 108.8%,115.6%,98.35%,140.4%,and 107.6%,respectively.Considering its remarkable ability,the S.cellulosae AU‐10 lipase can be considered as a potential additive in the detergent industry.
