摘要 目的为研究裸花紫珠正丁醇提取物抗大鼠宫颈炎的作用机制。方法为取雌性SD大鼠36只,随机分为空白组,模型组,裸花紫珠正丁醇提取物高、中、低剂量组,妇炎洁组,每组6只。取20%苯酚胶浆连续7天阴道给药,制备大鼠宫颈炎模型,然后分别用不同浓度的裸花紫珠正丁醇提取物、妇炎洁阴道连续给药14天,给药结束后采用心脏采血法取动物血液,用酶联免疫吸附法(ELISA)测定各组动物血浆肿瘤坏死因子(TNF-α),白细胞介素(IL-1)和前列腺素(PGE2)的含量,并取各组大鼠宫颈组织行病理形态学检查。结果显示,不同浓度的裸花紫珠提取物组和妇炎洁组与模型组相比较,均可降低其血浆PGE2、TNF-α的含量,升高血浆IL-1水平。组织病理形态学检查显示不同浓度的裸花紫珠提取物组和妇炎洁组可以减轻宫颈炎组织形态学的损伤。结论:裸花紫珠正丁醇提取物可能通过抑制组织中PGE2、TNF-α的合成,增强组织IL-1水平,从而提高机体免疫应答,影响炎性反应相关细胞因子的表达而治疗宫颈炎症。 Objective:To study the anti-cervicitis effect in rat of n-Butanol extract from callicarpa nudiflora.Methods:Thirty-six female SD rats were randomly divided into a blank group and a model group,a high-,medium-,and low-dose group of callicarpa nudiflora n-butanol extract,and Fuyanjie group,with 6 rats in each group.Rat model of cervicitis was prepared by vaginal administration of 20%phenolic glue for 7 days,and then continuously administered with different concentrations of callicarpa nudiflora n-butanol extract and FuyanJie vaginal for 14 days.Animal blood was taken by cardiac blood sampling,and the levels of plasma tumor necrosis factor(TNF-α),interleukin(IL-1)and prostaglandin(PGE2)in each group were measured by enzyme-linked immunosorbent assay(ELISA).Cervical tissues of each group was taken to perform the pathological examination.Results:Compared with the model group,the different concentrations of the callicarpa nudiflora extract group and the Fuyanjie group showed the reduction on the plasma PGE2,TNF-αcontent and increase on the plasma IL-1 level.Histopathological examination showed that different concentrations of callicarpa nudiflora extract group and Fuyanjie group can alleviate the morphological damage of cervicitis.Conclusion:N-butanol extract from callicarpa nudiflora may Enhance tissue IL-1 levels by inhibiting the synthesis of PGE2 and TNF-αin tissues,thereby increasing the body’s immune response and treating the cervicitis through affecting the expression of Inflammatory response cytokines.
